黑木耳多糖对B16黑色瘤细胞抗肿瘤作用研究 被引量:27

摘要 目的探讨黑木耳多糖对B16黑色瘤细胞抗肿瘤作用及相关可能的机制。方法以B16细胞为研究对象,检测不同浓度的黑木耳多糖对B16细胞的增殖、伤痕愈合率的影响;建立黑色素瘤模型,检测黑木耳多糖对荷瘤小鼠体质量、肿瘤大小的影响,利用RT-PCR技术检测其对肿瘤组织Bax、P53、Caspase-3mRNA表达的影响。结果黑木耳多糖在体外抑制细胞伤口愈合,对B16细胞的抑制率呈剂量依赖性增长,24 h、48 h、72 h的半数抑制浓度分别是80.99、43.18、70.82μg/mL。在体内,与模型组比较,灌胃黑木耳多糖能明显抑制肿瘤生长,且与环磷酰胺联合治疗,其抑制作用增加;黑木耳多糖可促进肿瘤组织中Bax、P53、Caspase-3 mRNA的表达。结论黑木耳多糖在体内、外都具有明显的抗肿瘤作用,这可能与其能促进肿瘤细胞凋亡有关。 Objective To investigate the antitumor effect of Auricularia auricula polysaccharides and its possible mechanism on B16 melanoma cells. Methods The proliferation and wound healing of B16 cells were detected after treated with different concentrations of polysaccharides from Auricularia auricula. The melanoma mouse model was established. The effects of Auricularia auricula polysaccharides on body weight and tumor size of tumor mice were measured. RT-PCR method was used to detect the expression of Bax,P53 and Caspase-3 mRNA in tumor tissues. Results The inhibitory rate of Auricularia auricula polysaccharides on B16 cells was increased in a dose-dependent manner, and the median inhibitory concentrations after incubation for 24 h,48 h and 72 h were 80.99,43.18,and 70.82 μg/mL,respectively.Compared with the model group,treatment with Auricularia auricula polysaccharides significantly inhibited the growth of the tumor and upregulated the expression of Bax,P53 and Caspase-3 mRNA in tumor tissues.Meantime,the inhibition effect of Auricularia auricula polysaccharides was increased when combined with cyclophosphamide. Conclusion Auricularia auricula polysaccharide has obvious antitumor effect in vitro and in vivo,and its antitumor mechanism may be related to promote tumor cell apoptosis.
