脑清微乳鼻腔给药对大鼠脑出血的作用研究

摘要 目的研究脑清微鼻腔给药对脑出血的作用及其机理。方法采用大鼠尾状核注入自体尾动脉血诱导脑出血模型,将大鼠随机分为假手术组、对照组和脑清微乳组,脑清微乳组给予脑清微乳0.60 mL/kg,2次/d;假手术组和对照组等体积给予0.9%生理盐水,给药方式均为滴鼻。于规定时间点观察各组大鼠脑细胞形态的变化,并测定脑组织含水量及血肿周围AQP-4、MMP-9、Caspase-3、Bcl-2的表达变化。结果与假手术组比较,对照组和脑清微乳组大鼠均出现神经功能缺损症状、脑水肿,4种蛋白的表达水平均有增加(P<0.05),说明造模成功。与对照组比较,脑清微乳组的神经缺损症状和脑组织含水量均有明显改善(P<0.05);脑清微乳组的AQP-4、MMP-9和Caspase-3的表达水平都有明显的下降(P<0.05),Bcl-2则显著增加(P<0.05)。结论脑清微乳能通过调节AQP-4、MMP-9的表达有效改善大鼠脑出血引起的脑水肿,并通过调节Caspase-3/Bcl-2的表达有效改善大鼠脑出血引起的细胞凋亡。 Objective To study the effect and mechanism of Nao-Qing microemulsion on intracerebral hemorrhage in rats. Methods The intracerebral hemorrhage( ICH) rat model was established by injection of tail arterial blood into its caudate nucleus. Then the rats were randomly divided into the sham-operation group,the control group and Nao-Qing microemulsion group. The behavior changes of ICH rats were observed,and the changes of cellular morphology of brain tissue were measured. The brain water content and the expression of AQP-4,MMP-9,Caspase-3 and Bcl-2 were determined. Results Compared with the sham-operation group,the neurological deficit, the brain water content and four kinds of protein were all increased in ICH rats( P〈0. 05). Compared with the control group,the neurological deficit and the brain water content were attenuated in ICH rats after Nao-Qing microemulsion treatment. Meantime,the levels of AQP-4,MMP-9 and Caspase-3 were decreased,but Bcl-2 was increased in Nao-Qing microemulsion group.Conclusion Nao-Qing microemulsion can effective prevent against ICH brain injury,which may be related to regulation of cerebral edema-associated AQP-4 and MMP-9 expression, and cell apoptosis-associated Caspase-3 and Bcl-2 expression.
