小檗碱联合奥沙利铂对肝癌细胞SMMC7721的抑制增殖作用 被引量:3

摘要 目的探讨小檗碱联合奥沙利铂对人肝癌细胞SMMC7721的作用。方法 MTS法检测小檗碱与奥沙利铂在单独及联合用药时对SMMC7721细胞的杀伤作用,结晶紫染色检测细胞的存活情况,Edu法检测其对SMMC7721细胞的增殖影响,Western-blot法检测凋亡相关蛋白cleaved caspase 3及cleavedPARP的表达。结果 (1)与单药处理组比较,小檗碱与奥沙利铂联合用药组细胞的存活率明显下降(P<0.05),具有明显的时间依赖性;结晶紫染色实验结果也有类似现象。(2)Edu染色实验结果表明,联合用药组细胞增殖明显减少。(3)联合用药组凋亡相关蛋白cleaved-PARP、cleaved caspase 3的表达水平均较单药组明显上调。结论小檗碱能增加奥沙利铂对SMMC7721细胞的杀伤作用,其机制可能与促进肝癌细胞凋亡有关。 Objective To explore the antitumor efficacy of berberine combined with oxaliplatin on hepatocellular carcinoma. Methods Cell viability was determined by MTS assay. Cell proliferation was measured by Edu cell proliferation kit. The expression of apoptosis-related proteins cleaved caspase 3 and cleaved PARP were measured by western blot. Results The combination of berberine and oxaliplatin exerted a synergistic inhibitory effect on the proliferation of SMMC7721 cells in a time-dependent manner.The Edu staining revealed that the combination of 50 μmol/L berberine and 3. 125 μmol/L oxaliplatin exhibited a significant anti-proliferation effect on SMMC7721 cells. Moreover, berberine combined with oxaliplatin obviously induced the expression of cleaved PARP and cleaved caspase-3. Conclusion The co-adminstration of berberine and oxaliplatin inhibits the growth of SMMC7721 cells,which may be related to promotion of cell apoptosis.
