摘要
白玉蜗牛的蛋白腺、肝脏、肌肉、血液经pH为7.4的磷酸缓冲液抽提,80%饱和硫酸铵沉淀粗分离,Q Sepharose HP强阴离子交换色谱及Superdex G-75葡聚糖凝胶色谱分离得到白玉蜗牛各组织的凝集素(Achatina fulica ssp.Lectin,AFL).血细胞凝集实验显示:AFL对兔血红细胞具有高凝集活性,其最小凝集质量浓度为0.065μg/mL;热处理15min后不同组织提取的AFL热稳定性不同,其中血液AFL在60℃处理后仍然具有较好的凝集活性.不同糖与不同组织提取的AFL对兔血红细胞的凝集抑制作用有明显差异,揭示了不同组织AFL可能与不同糖存在特异性结合.
The protein gland,liver,muscle and blood of snail Achatina fulica ssp.were extracted by phosphate buffer solution with pH 7.4,80% saturated ammonium sulfate precipitate,Q Sepharose HP strong anion exchange chromatography and Superdex G-75 dextran gel,AFL(Lectin of Snail Achatina fulica ssp.)was obtained by chromatographic separation.Blood cell agglutination test showed that AFL had high agglutination activity in rabbit erythrocytes with a minimum agglutination concentration of0.065μg/mL.After heat treatment for 15 min,AFL in different tissues showed certain tolerance to heat treatment,and AFL extracted from blood treatment at 60 ℃ showed good agglutination activity.The inhibitory effect of different saccharides and AFL extracted from different tissues on the aggregation of red blood cells in rabbits revealed that AFL might be specifically differentiated from different saccharides.
