摘要
目的:探讨大鼠哮喘气道重塑过程中不同阶段气管平滑肌细胞(ASMC)分泌的Ⅲ型胶原蛋白功能的变化及孟鲁司特对Ⅲ型胶原蛋白的影响。方法:应用卵蛋白致敏激发建立哮喘气道重塑大鼠模型,原代培养ASMC,通过免疫组织化学方法检测ASMC中Ⅲ型胶原蛋白的表达。结果:哮喘组Ⅲ型胶原蛋白平均灰度值均高于对照组,以卵蛋白激发8周组最高,差异有统计学意义(P均<0.01);孟鲁司特干预组Ⅲ型胶原蛋白平均灰度值明显低于哮喘气道重塑组,尤以孟鲁司特干预2周及4周组差异更显著(P<0.01)。结论:孟鲁司特通过抑制ASMC分泌的Ⅲ型胶原蛋白合成能力而减轻气道重塑。
Objective:To investigate the effect of montelukast on the secretion of type Ⅲ collagen in airway smooth muscle cells(ASMC) during different stages of airway remodeling in asthmatic rats.Methods:Ovalbumin(OVA) was used to establish asthmatic airway remodeling model of SD rats and montelukast was given to interfere.Primary culture purified ASMC,and measured the expression of type Ⅲ collagen in ASMC by immunochemical method.Results:The average gray value of type Ⅲ collagen in asthma group was higher than that of the control group,and OVA motivated 8 weeks group was the highest.The differences were significant(P〈0.01).The average gray value of type Ⅲ collagen in montelukast group was significantly lower than that in asthmatic airway remodeling group,it was significantly different in 2 weeks group and 4 weeks group(P〈0.01).Conclusion:Montelukast can reduce airway remodeling by inhibiting ASMC collagen synthesis ability.
