HER-2在结直肠癌组织中的表达及其临床意义

Expression of Human Epidermal Growth Factor-2 in Patients with Colorectal Cancer

目的研究人类表皮生长因子受体-2(human epidermal growth factor receptor 2,HER-2)在结直肠癌组织中的表达及特征。方法运用免疫组化的方法检测经外科手术切除的50例结直肠癌组织标本和30例正常结直肠组织标本中HER-2的表达状况,并研究HER-2表达与结直肠癌病理特征和临床表现的相关性。结果 HER-2在结直肠癌组织中表达阳性18例(36.0%),正常组织阳性3例(10.0%),2组差异有统计学意义(P<0.05)。HER-2基因表达与结直肠癌患者的年龄、部位及性别等无关(P>0.05),但是与结直肠癌的分期、患者淋巴结转移等有着明显的相关性(P<0.05)。HER-2阳性的结直肠癌患者5年生存率显著低于阴性患者(P<0.05)。结论结直肠癌患者中存在HER-2蛋白过表达,HER-2或成为结直肠癌的一种早期检测和诊断标志物。

Objective To investigate the expression of HER-2 in patients with colorectal cancer and the relationship be- tween its expression and patients. Methods Immunohistochemistry was used to detect the expression of HER-2 in 50 cases of colorectal cancer tissue and 30 cases of normal colorectal tissue. Pathological characteristics and clinical data were compared be- tween those two groups. Results HER-2 was positively expressed in 18 cases （36.0%） of colorectal cancer tissue and 3 cases （ 10.0% ） of normal tissue, and the difference was statistically significant （ P 〈 0.05 ）. The over expression of HER-2 was not as- sociated with patients, age, gender, tumor size, location and distant metastases （ P 〉 0.05 ）, but associated with differentiation stage, Dukes＇ stage and lymphatic metastases （ P 〈 0.05）. The colorectal cancer patients with positive expression of HER-2 had a significantly lower five-year survival than those with negative expression （P 〈 0. 05 ）. Conclusion The expression of HER-2 was associated with the occurrence of colorectal cancer, which could be used as a marker for early detection and prognosis.
