三拗汤及其拆方对OVA建立小鼠支气管哮喘模型血清Ig-E及BALF液IL-4、IL-5、IL-6的影响 被引量:9

摘要 目的:观察三拗汤对小鼠支气管哮喘变应性气道炎症细胞因子的影响。方法:Balb/c小鼠90只随机分为空白组,模型组,地塞米松阳性组(1 mg·kg^(-1)),三拗汤高剂量组(4.5 g·kg^(-1)),三拗汤低剂量组(2.25 g·kg^(-1)),麻黄杏仁高剂量组(3 g·kg^(-1)),麻黄杏仁低剂量组(1.5 g·kg^(-1)),麻黄甘草高剂量组(3 g·kg^(-1)),麻黄甘草低剂量组(1.5 g·kg^(-1))。各组小鼠(除空白组外)于第1天、第8天、第14天腹腔注射0.2 mL致敏液,0.2 mL致敏液含卵清蛋白(ovalbumin,OVA)0.1 mg、氢氧化铝0.02 mg。第15天—第29天(除空白组)给予质量分数5%的OVA雾化激发,每次激发20 min,每次激发前30 min各组小鼠按体质量给予相应的药物。空白组以等体积生理盐水代替致敏液,并用生理盐水激发相同时间。两周后,最后一次OVA雾化后摘眼球取血,ELISA法测定血清IgE;收集肺泡支气管灌洗液(bronchoalveolar lavage fluid,BALF),ELISA法检测白细胞介素-4(interleukin-4,IL-4)、IL-5、IL-6;取左肺做病理组织学观察。结果:模型组血清IgE及BALF液中IL-4,IL-5,IL-6含量显著增高。肺组织病理显示肺脏可见间质多灶性炎细胞浸润(++);肺泡壁轻度充血(+),肺泡壁明显增厚。三拗汤各组能降低血清IgE及BALF液中IL-4、IL-5、IL-6含量,减少上述模型的病理改变。结论:三拗汤对OVA导致的过敏性哮喘有明显抑制作用,可降低哮喘小鼠血清中IgE分泌及降低BALF液中IL-4、IL-5、IL-6水平,且效果优于各拆方组。 Objective: To evaluate the effect of San' ao Decoction on airway inflammatory cytokines in bronchial asthma in mice.Methods: Ninety Balb/c mice were randomly divided into blank group,model group,dexamethasone positive group( 1 mg ·kg^-1),San'ao Decoctdion high dose group( 4. 5 g·kg^-1),San'ao Decoctdion low dose group( 2. 25 g·kg^-1),Ephedra Almond high dose group( 3 g·kg^-1),Ephedra Almond low dose group( 1. 5 g·kg^-1),Ephedra Licorice high dose group( 3 g·kg^-1)and Ephedra Licorice low dose group( 1. 5 g·kg^-1). The mice in each group( except the blank group) were intraperitoneally injected with 0. 2 mL of sensitizing solution on day 1,day 8,and day 14,0. 1 mL of ovalbumin( OVA) 0. 2 mg,aluminum hydroxide 0. 02 mg. On the 15 th day to the 29 th day( excluding the blank group),5% OVA atomization was used to stimulate the mice.Each challenge 20 min,each group of mice was given corresponding drugs by body weight 30 min before each challenge. The blank group replaced the sensitized solution with an equal volume of normal saline and stimulated the same time with normal saline. Two weeks after the last OVA nebulization,blood was taken from the eyes and serum IgE was measured by ELISA. Bronchoalveolar lavage fluid( BALF) was collected and interleukin-4( IL-4),IL-5,IL-6; take left lung histopathological observation. Results: The contents of IL-4,IL-5 and IL-6 in serum of IgE group and BALF fluid in model group were significantly higher than those in control group. Pathological examination of lung showed interstitial multifocal inflammatory cell infiltration,mild hyperemia of the alveolar wall and thickening of the alveolar wall. San' ao Decoction can reduce the levels of IL-4,IL-5 and IL-6 in serum IgE and BALF,and reduce the pathological changes of the above models. Conclusion: San'ao Decoction can significantly inhibit the allergic asthma induced by OVA,reduce the secretion of Ig-E in serum and decrease the levels of IL-4,IL-5 and IL-6 in BALF The demolition of the group.
