摘要
以青海湖裸鲤体表为实验材料,应用平板划线法分离和纯化后得到一株稳定的淡水发光菌株,编号为FG1.该发光细菌为革兰氏阴性菌株,有鞭毛具有运动性.氧化酶、V-P试验、淀粉水解、甲基红等试验均表现出阳性反应,能发酵葡萄糖、蔗糖、丙三醇等产酸,最适生长pH为9.0~10.0,最适生长温度为25℃.16Sr DNA基因序列分析结果表明,与FG1菌株的16Sr DNA基因序列同源性最高的菌株为Vibrio anguillarum,相似率为100%,结合该菌株的形态学特征、生理生化特性及分子生物学特性,将FG1菌株鉴定为Vibrio anguillarum.
A luminous bacterium strain FG1 was isolated from the body surface of G. przewalskii by using flat line. The results showed that it was gram-negative strain and had flagellum and motility. Through detection of its physiological and biochemical characteristics such as ferment glucose,sucrose,glycerol,produce acid,oxidase,V-P test,secret amylase,and methyl red,all the test results showed positive reaction. Its optimum growth temperature was 25℃ and optimum growth pH was 9. 0 - 10. 0. In conclusion,it was identified as Vibrio anguillarum based on morphological,biochemical and physiological characteristics combined with analysis of 16 Sr DNA.
