臭氧水疗治疗变应性接触性皮炎样小鼠的疗效观察 被引量:4

摘要 目的观察臭氧水疗治疗变应性接触性皮炎样小鼠的疗效。方法 DNCB诱导的变应性接触性皮炎样小鼠模型40只,随机分成A组(臭氧水疗组),B组(药物治疗组),C组(纯净水治疗组),D组(自然恢复组)。A、B、C三组皮损处分别予以臭氧水疗、外用药物、纯净水治疗,连续治疗2周,D组不做任何处理。2周后处死小鼠,HE染色观察各组小鼠的皮肤组织病理改变;透射电镜观察各组小鼠皮肤棘层的变化;实时定量PCR(RT-PCR)检测皮损中γ-干扰素(IFN-γ)、白介素-10(IL-10)、白介素-17(IL-17)表达水平。结果 A组及B组较C组及D组小鼠的搔抓症状及皮炎表现好转,皮损处病理结果与电镜下超微结构明显改善。A组及B组小鼠皮损中IL-10、IL-17 m RNA水平明显低于C组和D组,IFN-γm RNA水平较C组及D组明显升高,差异有统计学意义(P<0.05)。结论臭氧水疗对治疗变应性接触性皮炎样小鼠有明显疗效,可能通过调控T细胞间的平衡及细胞的氧化还原反应发挥治疗作用。 Objective To observe the effect of ozone hydrotherapy on allergic contact dermatitis(ACD)mice.Methods Forty mice with ACD induced by dinitrochlorobenzene(DNCB)were randomly divided into four groups: group A(ozone hydrotherapy group), group B(drug therapy group), group C(purified water therapy group)and group D(natural recovery group).Skin lesions of groups A, B and C were treated with ozone liquid, topical drug, purified water respectively for 2 weeks, and the group D was received no treatment.Two weeks later, the mice were killed and the skin lesions were collected and the pathology of skin tissues of each group were observed with HE dyeing; the changes of skin spines in mice were observed by electron microscopy; the expression level of Interferon-γ(IFN-γ), Interleukin-10(IL-10)and Interleukin-17(IL-17)in the skin lesions were quantitatively detected by real-time PCR(RT-PCR).Results Scratching symptoms, pathological results and ultrastructural of skin lesions of mice in group A and group B were better than these in groups C and D.The expression levels of IL-10 and IL-17 mRNA in skin tissues of both group A and group B were clearly lower than those of the group C and the group D; on the contrary, the level of IFN-γ mRNA of the two groups were higher than those of the latter two groups(P〈0.05).Conclusion Ozone hydrotherapy has a significant effect on ACD mice through adjusting and controlling the imbalance of T cell and cell redox reaction.
