益生菌可改善高脂饮食诱导的雄性大鼠睾丸凋亡

摘要 目的探究益生菌在高脂饮食诱导的生精功能异常中的作用。方法健康成年雄性sD大鼠随机分为3组.包括正常对照组、高脂饮食组和益生菌组。正常对照组给予正常饲料喂养,高脂饮食组给予高脂饮食喂养,益生菌组在高脂饮食喂养的同时给予益生菌灌胃,8周后,检测3组大鼠生精功能的改变。结果高脂饮食组大鼠生精功能明显受损,表现为睾丸重量/体重下降,血清睾酮下降,睾丸HE染色出现病理性异常改变。同时,睾丸组织凋亡蛋白Fas表达增加,Bcl—xl表达下降。益生菌干预之后,大鼠生精功能得到明显改善,睾丸重量/体重增加[(8.98±0.43)/25.(6.90±0.50),P〈0.05],血清睾酮增加[(2.31±0.21)vs.(1.67±0.13),P〈0.05],睾丸组织形态学分析明显改善。与此同时,益生菌干预之后,睾丸组织凋亡蛋白Fas表达降低[(4072.50±105.83)vs.(4611.25±140.68),P〈0.05],Bcl—xl表达升高[(4047.50±210.92)掷.(3142.50±113.14),P〈0.05]。结论益生菌可改善高脂饮食诱导的雄性大鼠生精功能受损,减轻睾丸组织局部的细胞凋亡。 Objective To investigate the effects of probiotics on high-fat diet (HFD) -induced spermatogenesis dysfunction. Methods Healthy adult male rats were randomly divided into three groups, including control group, HFD group and probiotics group. Rats in the control group received common diet, rats in the HFD group were fed with HFD and rats in the probiotics group were given HFD feeding and oral probiotics. The spermatogenesis of the rats in the three groups was observed 8 weeks later. Results Spermatogenesis function of the male rats was destroyed by HFD feeding, manifested as pathological histological changes, decreased testis weight/body weight, and decreased serum testosterone level. The expression of Fas was increased in the testis of the HFD group and the expression of Bcl-xl was decreased by HFD feeding. Probiotics treatment obviously improved spermat- ogenesis function of the male rats, as assessed by improved histological morphology of testis, in- creased testis weight/body weight [ ( 8.98 ±0. 43 ) vs. (6.90 ± 0.50), P 〈 0. 05 ] and increased serum testosterone level [ (2. 31 ±0. 21 ) vs. ( 1.67 ± 0. 13 ), P 〈 0. 05 ) . What' s more, the Fas [ (4 072.50 ± 105.83) vs. (4 611.25 ±140. 68), P 〈0.05] was decreased and the Bcl-xl [ (4 047.50±210.92) vs. (3 142.50±113.14), P 〈0.05) was increased by probiotics treat- ment. Conclusion s The probiotics cas ameliorate HFD-induced spermatogenesis deficiency and re-duce cell apoptosis of testis.
