摘要 目的探讨显色原位杂交技术在石蜡切片中检测EBER的操作与常见问题分析。方法采用显色原位杂交技术,选取15例鼻咽黏膜慢性炎病理活检组织,25例鼻咽非角化鳞状细胞癌病理活检组织行EBER检测。结果 15例鼻咽黏膜慢性炎中有2例EBER呈点状阳性(阳性率13.3%),25例鼻咽非角化鳞状细胞癌全部阳性(阳性率100%)。结论采用显色原位杂交技术检测EBER,阳性信号定位准确,染色效果良好,无背景染色。杂交时间缩短,提高了检测效率,为EB病毒感染相关疾病的病理诊断及鉴别提供有效的依据。 Objective To investigate the operation and common problems of chromogenic in situ hybridization in detecting EBER in paraffin sections. Methods Using chromogenic in situ hybridization, 15 pathological tissues with chronic inflammation of nasopharyngeal mucosa, and 25 pathological tissues with nasopharyngeal non-keratinized squamous cell carcinoma were selected for the detection of EBER. Results Among the 15 cases of chronic inflammation of nasopharyngeal mucosa, 2 cases showed dot-distributedpositive EBER(positive rate, 13.3%). All 25 cases of nasopharyngeal nonkeratinized squamous cell carcinoma presented positive(positive rate, 100%). Conclusion Application of chromogenic in situ hybridization in detecting EBER shows accurate localization with positive signals, good effect in staining, and no background staining. The shortening of hybridization time improves detection efficiency and provides an effective basis for the pathological diagnosis and identification of EB-virus related diseases.
