浅谈辛伐他汀对大鼠脂肪源性干细胞的增殖和成骨分化的影响

摘要 目的研究分析大鼠脂肪源性干细胞的增殖及骨代谢受到辛伐他汀的影响。方法取大鼠腹股沟的脂肪,将脂肪肝细胞进行分离,然后使用贴壁法对纯化细胞进行筛选。选取第三代细胞,通过加入各浓度的辛伐他汀CCK-8来对细胞增殖能力进行分析,使用茜素红染色对骨诱导矿化情况进行分析。结果在接种24 h后,细胞体比较小,形状为星形,存在漂浮脂肪液滴。接种48 h之后,部分细胞出现集落生长,细胞的体积不断增加。接种72 h后,胞生长呈现纤维状,部分聚集成团。经过7天培养,梭形细胞快速生长,细胞密度达到80%,可以进行传代。随着辛伐他汀浓度的增加,ADSCs能力降低,1μm/L组与对照组的ADSCs增殖能力无差异性,10μm/L组对ADCSs的增殖有明显的抑制作用。CCK-8结果显示,10μm/L浓度的辛伐他汀对ADSCs的增殖有明显的抑制作用。结论大鼠脂肪源性干细胞增殖和成骨分化会受到辛伐他汀浓度的影响。 Objective To investigate the effects of simvastatin on the proliferation and bone metabolism of rat adipose derived stem cells. Methods The fat of the rat's groin was taken and the fatty liver cells were separated. Then the purified cells were screened by the method of adherence. Third generation cells were selected to analyze cell proliferation ability by adding various concentrations of simvastatin CCK-8, and the mineralization of bone was analyzed by alizarin red staining. Results After 24 hours of inoculation, the cell body was smaller and the shape was star shaped, and there was floating fat droplet. After 48 hours of inoculation, colony growth was found in some cells, and the volume of cells increased. After 72 hours of inoculation, the cells grew fibrous and partially aggregated into groups.After seven days of culture, the spindle cells grew rapidly and the cell density reached 80%, which could be carried out. With the increase of simvastatin concentration, the ability of ADSCs decreased. There was no difference in the proliferative capacity of ADSCs between 1 μm/L group and control group. The 10 μm/L group significantly inhibited the proliferation of ADCSs. CCK-8 results showed that simvastatin with a concentration of 10 μm/L had a significant inhibitory effect on the proliferation of ADSCs. Conclusion The proliferation and osteogenic differentiation of adipose derived stem cells in rats are affected by the concentration of simvastatin.
