Determination of gentamycin sulfat in urine by room-temperature phosphorescence sensor based on Mn-ZnS quantum dots

摘要 目的:基于硫酸庆大霉素对Mn掺杂ZnS量子点的磷光增敏作用,建立一种简便且灵敏测定硫酸庆大霉素浓度的新方法。方法:在水相中合成巯基丙酸(MPA)包裹的Mn掺杂ZnS量子点,实验中考察了缓冲溶液pH和类型、反应时间及量子点浓度等因素对室温磷光检测的影响。利用荧光分光光度计通过测定Mn掺杂ZnS量子点磷光强度的变化,实现对尿液中硫酸庆大霉素以及硫酸庆大霉素注射液浓度的测定。结果:成功合成了浓度为2×10^(-2)mg/L的Mn掺杂ZnS量子点。在pH=7.4的三羟甲基氨基甲烷-盐酸缓冲体系中,静置反应时间为30min条件下,硫酸庆大霉素浓度与Mn掺杂ZnS量子点磷光强度线性关系良好且相关系数R^2=0.9985。结论:该方法灵敏、快捷,可以用于测定尿液中硫酸庆大霉素的浓度。 Objective: To establish a simple and sensitive method for the determination of gentamicin sulfate based on the photosensitization of Mn-Zn S quantum dots. Methods: propionic acid(MPA) modified Mn-Zn S quantum dots were synthesized in aqueous phase. The effects of pH,type of buffer solution, reaction time and concentration of quantum dots on the room-temperature phosphorescent detection were investigated.The concentrations of gentamicin sulfate injection and gentamicin sulfate in urine were determined by measuring the phosphorescence intensity of Mn-Zn S quantum dots with a fluorescence spectrophotometer. Results: The Mn-Zn S quantum dots were successfully synthesized. The linear relationship between gentamicin sulfate and phosphorescence intensity of the Mn-Zn S quantum dots(2 ×10^-2 mg/L) was obtained with a correlation of 0.998 5 after 30 min incubation in Tris-HCl buffer(pH =7.4) at room temperature. Conclusion:The proposed method is sensitive and simple and can be used for the determination of gentamicin sulfate in urine.
