卵泡抑素样蛋白1与糖尿病动脉粥样硬化的炎症反应 被引量:1

Follistatin-like 1 and inflammatory responses of diabetic atherosclerosis

摘要 目的:探究细胞因子卵泡抑素样蛋白1(FSTL1)参与调节巨噬细胞促进糖尿病动脉粥样硬化斑块形成的机制。方法:采用酶联免疫吸附测定(ELISA)方法检测动脉粥样硬化患者和糖尿病动脉粥样硬化患者血清FSTL1表达水平。构建糖尿病小鼠模型,利用免疫组织化学(组化)检测FSTL1表达阳性区域;使用氧化型低密度脂蛋白(ox LDL)和(或)FSTL1刺激人巨噬细胞后,采用ELISA方法检测其炎症因子的分泌水平,探讨FSTL1参与糖尿病动脉粥样硬化发生、发展的机制。结果:细胞因子FSTL1在糖尿病动脉粥样硬化病变中表达异常升高,糖尿病动脉粥样硬化组为(13.60±4.20)μg/L,动脉粥样硬化组为(9.90±3.50)μg/L(P<0.01),且主要表达于巨噬细胞;FSTL1可刺激巨噬细胞的炎症因子释放,触发局部炎症反应。结论:细胞因子FSTL1可能通过激活巨噬细胞炎症反应,参与糖尿病动脉粥样硬化斑块的发生、发展。 Objective To investigate the mechanism of the cytokine follistatin-like 1( FSTL1) in regulating macrophages to promote plaque formation in diabetic atherosclerosis. Methods To detect the expression level of FSTL1 in serum of patients with atherosclerosis and diabetic atherosclerosis by enzyme-linked immunosorbent assay(ELISA). A diabetic mouse model was established and detect the positive area of FSTL1 expression immunohistochemically. The levels of inflammatory cytokines in human macrophages were detected by ELISA after stimulating human macrophages by using oxi-dized low density lipoprotein(ox LDL) or/and FSTL1, and the mechanism of FSTL1 in the development of diabetic atherosclerosis was explored. Results The expression of cytokine FSTL1 was abnormally elevated in diabetic atherosclero-sis than in atherosclerosis [(13.60±4.20) μg/L vs(9.90±3.50) μg/L(P0.01)], and it was mainly located in macrophages.FSTL1 stimulated the release of inflammatory cytokines in macrophages and triggered local inflammatory responses.Conclusions The cytokine FSTL1 may be involved in plaque progression in diabetic atherosclerosis through activating in-flammatory responses in macrophages.
