卵泡期和黄体期牦牛输卵管中Bcl-2和Bax的表达变化 被引量:2

Difference of Bcl-2 and Bax expression in the yak oviduct during follicular and luteal phases

摘要 为了研究发情周期不同阶段牦牛输卵管中Bcl-2和Bax的表达变化,采用免疫组织化学SP法和实时荧光定量聚合酶链式反应(real-time fluorescent quantitative PCR,q RT-PCR)技术分别对黄体期和卵泡期牦牛输卵管中Bcl-2和Bax蛋白及其m RNA的表达特征进行了研究。结果显示,发情周期不同阶段Bcl-2和Bax蛋白免疫阳性产物主要表达于牦牛输卵管黏膜上皮细胞质中,且主要分布于上皮细胞游离面和基底面。黄体期Bax蛋白阳性产物表达量显著高于卵泡期,而Bcl-2表达量略低于卵泡期,但无显著差异。卵泡期和黄体期牦牛输卵管管壁中有Bcl-2和Bax m RNA表达,不同时期Bcl-2 m RNA变化无显著差异;黄体期Bax m RNA表达显著高于卵泡期。表明,Bcl-2和Bax参与了牦牛输卵管黏膜上皮细胞周期性增殖与凋亡的调控,而且这一凋亡通路中主要通过Bax的变化来实现。 The methods of Streptavidin-Perosidase(SP) immunohistochemistry and real-time flu- orescent quantitative PCR were used to study the expression of Bcl-2 and Bax in the yak oviduct in a fol- licular and luteal phase. The results showed that the Bcl-2 and Bax proteins were expressed in the cyto- plasm of the oviduct epithelium, and their immunoreactivity was mainly detected in the free and basal surface of the epithelial ceils. In the luteal phase,Bax immunoreactivity was significant higher than that in the follicular phase, but Bcl-2 immunoreactivity in the luteal phase were decreased than that in the follicular phase with no significance. In follicular and luteal phases,Bcl-2 and Bax mRNAwere expressed in the yak oviduct,and Bcl-2 expression had no significant difference in follicular and luteal phas s,but at the luteal phases,Bax was significantly higher than that in the follicular phase.The results suggested that Bcl-2 and Bax are involved in the regulation of cyclic cell apoptosis of the yak oviduct epithelium,and for this apoptosis pathway,it takes effects mainly through the regulation of Bax expression.
