摘要
本文通过对辽宁绒山羊组织DNA的提取、DNA浓度和纯度的检测、基因目的片段的扩张、序列测定与SNP定位等试验,得到了micro RNA的分离与靶标结合的特点,筛选得到的micro RNA的特点等。试验结果表明,利用在线软件mi RBase查找到与羊相关的micro RNA有137个相关,用Segal Lab of Computational Biology软件将ESR基因与羊的micro RNA结合情况分析后,发现突变前结合能够与靶基因结合的micro RNA有2个,突变后结合的micro RNA也是2个。筛选得到的RNA测序后通过基因序列与波峰的比对发现,在ESR基因3′-UTR区发现了1个差异位点,648 bp位点A/T的突变,旨在为今后羊的多态性的研究提供科学的理论依据。

This article through to the ESR gene 3'-UTR region mutation and the combination of micro RNA analysis,screening out the relevant micro RNA, aimed at the future of the study of the polymorphism of cashmere goats to provide a scientific basis for the theory, provide a theoretical basis for further study of animal micro RNA.
