摘要
单细胞测序技术已经应用在多个医学领域,在揭示细胞的异质性、环境中无法培养的微生物遗传信息和微生物的变异性等方面具有独特优势,能更深入层次的揭示生命活动的本质和规律。本文概括了当前单细胞分离技术、单细胞测序在基因组、转录组和表观遗传中涉及方法以及单细胞测序技术在病原微生物领域中的应用。
Single cell sequencing(SCS)has emerged as a valuable tool in diverse fields in recent years and it has become a hot topic in life sciences.SCS has many advantages in terms of studying cellular heterogeneity,examining the genetics of microbes in situations where culturing is not possible,and examining the variability of microbes.SCS offers a more in-depth look into the nature and regularity of biological activity.SCS has been used clinically in various fields,such as oncology,embryonic development,immunology,microbiology,and prenatal diagnosis.This review discusses advances in single cell manipulation,it summarizes the development of SCS methods in genomics,transcriptomics,and epigenomics,and it highlights the latest studies on pathogenic microbes.
