蔷薇红景天对大鼠脑缺血的保护作用研究 被引量:3

摘要 为观察蔷薇红景天对大鼠局灶性脑缺血模型的保护作用。采用Longa改良线栓法建立大鼠左侧大脑中动脉阻塞(MCAO)模型,通过观察大鼠脑组织病理切片,免疫组化,脑指数等指标,以及测定MDA、NO、GSH-PX、LD、Na+K+-ATP酶、Ca2+Mg2+-ATP酶的含量和SOD、NOS的活性,判断大鼠脑缺血情况及蔷薇红景天对大鼠脑缺血模型的保护作用,利用分子对接技术进一步阐述蔷薇红景天对大鼠脑缺血保护作用的机制。结果显示,蔷薇红景天能够显著降低大鼠血清中的MDA、NO、NOS和脑组织中LD的含量;显著升高血清中的SOD及脑组织中的GSH-PX、Na+K+-ATP酶、Ca2+Mg2+-ATP酶的含量;与模型组相比,各给药组的脑指数均有显著降低,从HE染色和免疫组化的图片来看,各给药组的脑缺血情况都有所改善,从分子对接的结果来看,主要是通过蔷薇红景天中化合物3、6、9、11、13、14对大鼠脑缺血起保护作用。由此可知,蔷薇红景天对大鼠局灶性脑缺血模型有一定的保护作用。 To investigate the protection of Rhodiola rosea L. on focal cerebral ischemic injury in rats. The middle cerebral artery occlusion(MCAO) in rats was adopted to establish cerebral ischemia model by Longa methods, which was validated by observing the histopathology slide, immunohistochemical markers and determining MDA, SOD, NO, NOS, GSH-PX, LD,Na+K+-ATPase and Ca2+-Mg2+ATPase. The mechanism of protective effect of Rhodiola rosea L. on cerebral ischemia in rats was further studied by using molecular docking technique. The results showed that Rhodiola rosea L. Significantly decreased the content of MDA, NO in serum of rats and LD in brain tissue of rats; The level of NOS and SOD was significantly increased in the serum tissue of rats and significantly increased the content of GSH-PX, Na+K+-ATPase and Ca2+Mg2+-ATPase in brain tissue of rats. Compared with the model group, the brain index of each treatment group was significantly lower;By HE staining and immunohistochemistry, there was a significant improvement in the degree of cerebral damage in the other groups compared with model group. It can be concluded that the Rhodiola rosea L. has some kind of protective effect on the focal cerebral ischemic injury in rats. From the result of molecular docking, the protective effects of compound 3, 6, 9, 13, 11 and 14 on cerebral ischemia are observed in Rhodiola rosea L.
