摘要
目的观察腓肠肌切口痛模型大鼠脊髓水平血管紧张素(AngⅡ)及其1型受体(AT1R)表达情况。方法健康成年SD大鼠16只随机分为腓肠肌切口痛模型组和对照组,分别于建模前,建模后3 h、1 d、2 d及3 d进行机械痛阈和热痛阈测定。完成行为学测定后处死大鼠取腰膨大处用脊髓ELISA法检测AngⅡ表达;免疫组织化学检测AT1R表达情况。结果切口痛模型组热痛阈与机械痛阈下降表现出明显的术后痛(P<0.01);与对照组比较,脊髓AngⅡ含量上升(P<0.01);正常大鼠脊髓组织有AT1R表达,腓肠肌切口痛模型组大鼠AT1R表达较正常对照组增多(P<0.01)。结论腓肠肌切口痛大鼠脊髓水平AngⅡ上升,AT1R表达增加。
Objective To evaluate the changes of angiotensin II and it’s type I receptor in spinal cord of a rat model of gastrocnemius incisional pain. Methods Sixteen Sprague-Dawley rats were randomly divided into gastrocnemius incision pain model group and sham group. Mechanical paw withdrawal threshold to von frey filament stimulation( MWT) and potential thermal withdraw latency ( PTWL) were measured prior to incision and at 3 h and 1, 2 and 3 days after incision. After behavior measurement,the rats were then euthanized the concentration of angiotensin II in spinal cord were measured by ELISAmethod and type 1 receptor of angiotensin II in spinal dorsal root ganglion were checked by immunohistochemistry. Results Compared with the baseline value before incision and sham group,MWT andPTWL were significantly decreased at 3 h and 1, 2 and 3 days after incision (P 〈 0 . 01 ) in study group. The con% in spinal of group incision was higher than that in sham group ( P 〈 0. 01) . Angiotensin % type 1 receptor of in spinal dorsal root gan-glion was higher in incision group than that in sham group ( P 〈 0. 01) . Conclusion Rats witli gastrocnemius incision had a higher con-centration of angiotensin % in spinal level and higher expression of angiotensin % type 1 receptor of in spinal dorsal root ganglion.[
