摘要
目的探究曲美他嗪对心肌缺血大鼠运动耐量的影响。方法研究时间为2017年6—8月,将冠脉结扎术后4周的30只SD大鼠随机分为模型组、小剂量曲美他嗪[20 mg/(kg·d)]组、大剂量曲美他嗪[40 mg/(kg·d)组,另设假手术组10只。灌胃给药4周,对比4组大鼠在治疗前、治疗2周、治疗4周时的运动耐量。结果治疗前、治疗2周、治疗4周时模型组大鼠运动耐量为(226.4±27.3)、(231.9±29.6)、(228.5±19.4)m,小剂量组为(221.7±25.9)、(257.8±26.4)、(304.1±35.8)m,大剂量组为(224.8±23.6)、(262.5±29.8)、(305.6±38.1)m,假手术组为(391.8±41.6)、(422.7±39.8)、(463.5±58.2)m,治疗前假手术组大鼠的运动耐量显著高于其他3组(P<0.05),模型组、小剂量组、大剂量组之间对比差异无统计学意义(P>0.05);治疗2、4周时假手术组大鼠的运动耐量仍显著高于其他3组(P<0.05),小剂量组、大剂量组大鼠运动耐量显著改善,且高于模型组(P<0.05),但小剂量组和大剂量组之间对比差异无统计学意义(P>0.05)。结论曲美他嗪能够有效提高心肌缺血大鼠的运动耐量,但不同剂量对大鼠运动耐量的影响并无明显差异。
Objective This paper tries to investigate the effect of trimetazidine on exercise tolerance in rats with myocardial ischemia. Methods 30 SD rats from June to August 2017 with coronary ligation after 4 weeks were randomly divided into the model group, low dose trimetazidine group [20 mg/(kg·d)], high dose trimetazidine group [40 mg/(kg·d)], another 10 of sham operation group. After intragastric administration for 4 weeks, the exercise tolerance of rats in the 4 groups was compared before the treatment and 2 weeks after treatment and 4 weeks after treatment. Results The exercise tolerance before treatment, 2 weeks and 4 weeks after treatment was(226.4±27.3),(231.9±29.6),(228.5±19.4)m, small dose group of(221.7±25.9),(257.8±26.4), 304.1±35.8)m, high dose group of(224.8±23.6),(262.5±29.8),(305.6±38.1)m; the sham operation group of(391.8±41.6),(422.7±39.8),(463.5±58.2)m, the exercise tolerance in the sham operation group before treatment was significantly higher than that of the other three groups(P〉0.05), no significant difference existed among model group, low dose group, high dose group(P〉0.05); for 2 weeks and 4 weeks, the exercise tolerance in the sham operation group was significantly higher than the other three groups(P〉0.05); the exercise tolerance in the low dose group and high dose group were significantly improved, and higher than the model group(P0.05), but no significant differences between low dose group and high dose group(P0.05). Conclusion Trimetazidine can improve the exercise tolerance of rats with myocardial ischemia,but there is no significant difference in the effect of different doses on exercise tolerance in rats.
