摘要
为了研究葡萄miR159基因家族在葡萄生长发育过程中发挥的作用,利用生物信息学工具和方法,对葡萄miR159家族成员进行了基因定位、前体序列二级结构预测、成熟序列保守性分析、靶基因及其功能预测以及系谱进化分析。结果显示:在葡萄miR159家族成员中vvi-miR159a和vvi-miR159b两个成员均分布在染色体15上,vvi-miR159c分布在染色体17上;葡萄miR159家族成员基本上符合进化规律;葡萄miR159家族成员的前体均可形成稳定的茎环结构,且其成熟序列都产生于其前体茎环结构的5'端臂上,其碱基序列保守性较高;葡萄miR159家族对应的靶基因主要有两个(GSVIVT01012447001与GSVIVT01037362001),经BLAST分析,主要为GAMYB转录因子家族。
In order to understand the functions of miR159 gene family in the growth and development of grape(Vitis vinifera),the author used the bioinformatics tools and methods to analyze or predict the gene mapping,the secondary structure of precursor sequences,the conservatism of mature sequences,the target genes and their functions,and the phylogenetic tree of miR159 family members in grape. The results showed that: among the members of miR159 family in grape,both vvi-miR159 a and vvi-miR159 b were distributed on chromosome 15,while vvi-miR159 c was distributed on chromosome 17. The phylogenetic analysis indicated that the miR159 family members in grape accorded with the evolution law basically. The precursors of miR159 family members in grape all could form stable secondary stem-loop structure,their mature sequences all were generated at 5' end arm of the stem-loop structure of precursors,and the bases of mature sequences were highly conserved. It was predicted that the target genes of miR159 family in grape mainly included GSVIVT01012447001 and GSVIVT01037362001,which mainly belonged to GAMYB transcription-factor family by BLAST analysis.
