摘要 目的分析血清样本保存温度、时间、密封状态、反复冻融、解冻温度和解冻时间对谷氨酸氨基转移酶(ALT)检测结果的影响。方法选取40例临床血清标本,按照不同方式处理后测定ALT浓度,并与初次检验值进行比较。结果血清标本密封状态保存下,于17~28℃保存≤1 d;2~8℃保存≤3 d;-20℃保存≤10 d;-80℃保存≤28 d,对ALT检测结果无明显影响。血清非密封状态保存下,17~28℃保存≤2 h;2~8℃保存≤8 h,对ALT检测结果无明显影响;从-20℃冰箱转至2~8℃下解冻≤90 min或转至17~28℃下解冻≤60 min;17~28℃、2~8℃反复冻融5次,对ALT检测结果均无明显影响。结论保存温度、时间、密封状态、解冻方式对血清样本检测影响显著,血液采集后应尽快检测,如需保存则要选择低温密封环境。 Objective To study the effects of serum samples store time, temperature, sealing, repeated freezing and thawing, thawing temperature and time on the results of analyzed glutamate aminotransferase(ALT) detection. Methods 40 clinical serum samples were selected, ALT concentrations were determined after treatment in different ways and compared with the initial values. Results It is showed that the serum samples preserved in sealed state, storage time is less than or equal to 1 day under the temperature from 17 ℃ to28 ℃, storage time is less than or equal to 3 days under the temperature from 2 ℃ to 8 ℃, storage time is less than or equal to 10 days under the temperature of minus 20 ℃, storage time is less than or equal to 28 days under the temperature of minus 80 ℃, there is no significant effect on ALT test results. The serum samples preserved in unsealed state, storage time is less than or equal to 2 hours under the temperature from 17 ℃ to 28 ℃, storage time is less than or equal to 8 hours under the temperature from 2℃ to 8 ℃, there is no significant effect on ALT test results. Thawing time is less than or equal to 90 minutes by transferring the serum from refrigerator of minus20 ℃ to the environment from 2 ℃ to 8 ℃, thawing time is less than or equal to 60 minutes by transferring the serum from refrigerator of minus 20 ℃ to the environment from 17 ℃ to 28 ℃. When the serum is frozen and thawed 5 times repeatedly, there is no significant effect on ALT test results. Conclusion The effect of temperature and storage time on serum sample detection is significant. Blood samples should be detected as soon as possible after the collection, and cryogenic sealed environment should be chosen if it needs to be preserved.
