基于均匀设计的复方金银花中药漱口水有效组分的配伍研究 被引量:9

摘要 目的:筛选茵陈(X_1)、野菊花(X_2)、金银花(X_3)、白芷及细辛(X_4)在复方金银花中药漱口水中的最佳配伍。方法:利用均匀设计的方法,并根据U8(8~5)均匀设计表,以茵陈、野菊花、金银花和细辛+白芷为因素,以各药物8个浓度为水平,以对牙龈卟啉单胞菌(Pg)的抑菌活性、生物膜形成抑制率及牙龈成纤维细胞毒性作用为指标,筛选5种药物的最佳配伍。结果:各复方对Pg的生长及生物膜形成均具有抑制作用。将数据进行二项式逐步回归,得出回归方程为Y=-0.9918-0.0044X_2-0.0002 X_1X_2-0.0005 X_1X_3+0.0004 X_1X_4+0.0017 X_2X_3;对回归方程进行优化后,获得各个因素的最佳配伍为:茵陈55 mg/mL,菊花45 mg/mL,金银花40 mg/mL,白芷+细辛(33∶17)40 mg/mL。结论:以茵陈、野菊花、金银花、白芷及细辛为有效成分的复方能有效抑制Pg,其最佳配伍为茵陈55 mg/mL,菊花45 mg/mL,金银花40 mg/mL,白芷+细辛(33∶17)40 mg/mL。 AIM:To sift the optimal ratio of artemisia capillaris thunb(ACT,X_1),chrysanthemum(CHR,X_2),honeysuckle(HDN,X_3),angelica dahurica and asarum sieboldii(AD and AS,X_4)by uniform design.METH-ODS:According to the uniform design and the U8(8~5)uniform design table,ACT,CHR,HON,AD and AS were used as the factors and 8 concentrations of each medicine as the levels.By evaluating the antibacterial activity against Porphyromonas gingivalis(P.gingivalis),the inhibition rate of biofilm formation and the toxicity on human gingival fibroblasts were explored,the optimal ratio of the 5 drugs was sieved.RESULTS:The above-mentioned compounds all showed inhibitory effects on the growth and biofilm formation of P.gingivalis.Binomial regression was applied to obtain the opti-mal ratio of the 5 drugs and the equation was Y=-0.9918-0.0044X_2-0.0002X_1X_2-0.0005X_1X_3+0.0004X_1X_4+0.0017X_2X_3.In addition,the optimal ratio was 55 mg/mL ACT,45 mg/mL for,40 mg/mL HON,40 mg/mL for ADand AS(33∶17).CONCLUSION:In the compound of honeysuckle traditional chinese medicine mouthwash the optimal ratio is 55 mg/mL for ACT,45 mg/mL for CHR,40 mg/mL for HON,40 mg/mL for AD and AS(33∶17).
