摘要 目的:从薏苡仁谷蛋白中获得高活性ACE抑制肽。方法:首先胃蛋白酶水解薏苡仁谷蛋白,进而采用超滤、离子交换色谱和反相高效液相色谱对其进行分离纯化,以体外ACE抑制活性为筛选指标,获得具有较高活性的多肽组分。结果:最终获得组分B2在浓度0.01 mg/m L时,ACE抑制率为(73.41±1.05)%。结论:薏苡仁谷蛋白经胃蛋白酶消化后,部分组分具有较高的降压潜力,该研究为中医药源降压药的开发及该药的临床应用提供新的理论数据。 To obtain high active ACE(Angiotensin Converting Enzyme) inhibitory peptides from glutelin of Coix seed.Methods: Glutelin of Coix seed was firstly hydrolyzed by pepsin,and the hydrolysate was then separated and purified using the ultrafiltration,ion exchange chromatography and RP-HPLC. Finally,the target peptide fraction was obtained under the guidance of ACE inhibitory activity in vitro. Results: Fraction B2 with higher activity was obtained and the inhibitory rate was 73. 41% ± 1. 05% at the concentration of 0. 01 mg/m L. Conclusion: Part components of the glutelin of Coix seed has high potential of anti-hypertensive effect after hydrolysis by pepsin,which provided guidance for further research and development of new peptide drugs from Chinese medicine,to provide new evidence for new application of Coix seed.
