摘要
【目的】对云耳菌丝进行耐高温比较及高温锻炼,以期获得相对耐高温的种质资源,为进一步选育适合广西高温地区栽培的优良云耳品种奠定基础。【方法】以菌丝生长速度为测定指标,结合菌落生长情况描述,对采集自广西田林、天峨等地的野生云耳分离菌株及广西目前主栽品种916进行不同生长温度、40℃下培养不同时间后的菌丝恢复能力及高温锻炼(40℃培养5 d,28℃培养7 d,42℃培养3 d,28℃培养7 d,45℃培养3 h,28℃培养7 d)后的菌丝生长情况比较。【结果】在高温区(31~37℃)菌丝生长速度及菌落生长较好的菌株为:HME2、TY026、YE1、TY041、YE2、YE(b);经40℃培养不同天数后,菌丝恢复较好的菌株为:YE1、TY071、TY041、TY026、HME2;经高温锻炼处理后,在28℃条件下培养仍能很好生长的菌株为:TY041、TY026、TY071、HME2、TY073。【结论】初步鉴定云耳菌株TY026、TY041和HME2为菌丝体耐高温品种,TY071和YE1耐高温性较好。
【Objective】High temperature resistance ability and heat acclimation of Auricularia auricula-judae mycelium were tested,in order to obtain high temperature resistant germplasm resources of Auricularia auricula-judae and provide breeding basis for cultivars of Auricularia auricula-judae suitable for cultivation in high temperature region in Guangxi.【Method】15 strains( including cultivar 916) of wide Auricularia auricula-judae collected from Tianlin and Tian'e county of Guangxi were studied by the following methods: Mycelium growth states in different temperature; Mycelium restorability testing after cultivated at 40 ℃ for different time( 3,5,7,and 9 days); Mycelium growth states after heat acclimation( process: 40 ℃ for 5 days,28 ℃ for 7 days,42 ℃ for 3 days,28 ℃ for 7 days,45 ℃ for 3 hours,28 ℃ for 7 days). 【Result】There were 6 strains[strain HME2,TY026,YE1,TY041,YE2,YE( b) ]which can grow well at the temperature of 31℃ to 37 ℃. After cultivated at 40 ℃ for different time,the mycelium restorability of 5 strains( YE1,TY071,TY041,TY026 and HME2) were better than other strains. After the heat acclimation,there were 5 strains( TY041,TY026,TY071,HME2,TY073) could grow well at 28 ℃.【Conclusion】Strains TY026,TY041 and HME2 were identified as mycelia high temperature resistance varieties,Y071 and YE1 have good heat resistance.
