摘要
目的分析高糖、高胰岛素对乳腺癌MCF-7细胞脂肪酸合成酶表达的影响及二甲双胍的干预效果。方法选择2009年5月~2015年5月在我院接受治疗的乳腺癌合并糖尿病患者78例,经病理活检、培育、检测后,分析检测结果。结果在高糖及高胰岛素状态下,MCF细胞脂肪酸合成酶在观察组体内的表达情况明显低于对照组,差异有统计学意义(P<0.05);经不同浓度培养观察组患者癌性病灶组织后,当浸入浓度20mmol/L的乳腺癌组织至少72h时,MCF-7的G1时期表达情况升高,而S及G2期表达情况则下降。结论在高糖及高胰岛素状态下,乳腺癌患者MCF-7细胞中存在的AMPK减少表达,G1期细胞减少,而二甲双胍可促使此状态下的AMPK增加表达,而且细胞成长过程暂停于G1期。
Objective To investigate the effect of high glucose and high insulin on expression of fatty acid synthase in MCF-7 cells of breast cancer and the intervention of metformin. Methods 78 patients with breast cancer and diabetes mellitus were selected from May 2009 to May 2015 in our hospital.After pathological biopsy,cultivation and detection,the results were analyzed.After pathologic biopsy,nurturing and testing,the results were analyzed. Results In high glucose and high insulin status,the expression of MCF cell fatty acid synthase in the observation group was significantly lower than that in the control group (P 〈0.05).After the tumor tissue in the observation group was cultured at different concentrations,the expression of MCF-7 in the G1 phase was increased when the breast cancer tissue immersed in the concentration of 20mmol/L at least 72h,while the expression was decreased in S and G2 phases. Conclusion In high glucose and high insulin condition,the expression of AMPK in MCF-7 cells in breast cancer patients decreased and G1 phase cells decreased,while metformin can increase the expression of AMPK in this state,and the cell growth process is suspended in the G1 phase.
