桑叶总黄酮提取工艺优化及其抗氧化活性分析 被引量:5

摘要 以桑叶总黄酮提取率为检测指标,通过单因素筛选及正交试验对桑叶总黄酮提取工艺进行了优化;并通过检测分析桑叶总黄酮清除ABTS^+·和DPPH·自由基能力来研究不同桑树品种的桑叶总黄酮体外总抗氧化活性。研究结果显示:桑叶总黄酮的最佳提取工艺为:提取时间120min,提取溶剂乙醇质量分数70%,料液质量浓度30g/L,提取温度80℃,优化后的桑叶总黄酮提取率达到33.97mg/g;不同桑品种的桑叶总黄酮提取液均对自由基ABTS^+·和DPPH·有清除能力,但不同品种间的清除率有极显著差异(P<0.01),其中昌盛、湘桑7920、佛堂瓦桑、鲁插1号有较强的ABTS+·清除能力,清除率在90%以上,皖桑2号、鲁插1号、湖桑199有较强的DPPH·清除能力,清除率在90%以上,表明桑叶总黄酮可作为一种良好的天然抗氧化剂来源。 With mulberry leaf flavonoids extraction rate as test indexes,the extraction process of mulberry leaf flavonoids was optimized by single-factor and orthogonal experiments.The total antioxidative capacity of mulberry leaf flavonoids of different mulberry varieties were studied according to detection of the ability of scavenging ABTS^+·and DPPH·free radicals.The results showed that:the optimum extraction technology of total flavonoids of mulberry leaves as follows,extraction time 120 min,extraction solvent ethanol concentration 70%,mass concentration of material liquid 30 g/L,extraction temperature 80 ℃.The ability of scavenging ABTS~+· and DPPH · free radicals of mulberry leaf flavonoids between different mulberry varieties had significant difference(P0.1).Changsheng,Husang 7920,Fotangwasang,Lucha 1 had higher scavenging capacity on ABTS+· radical with clearance rate more than 90%,and Wansang 2,Lucha 1,Husang 199 had strong scavenging capacity on DPPH ·radical with clearance rate above 90%.
