摘要
二甲基亚砜(DMSO)可直接作为药物用于抗炎、止痛等,还可因其特性作为诱导剂、有机溶剂、低温冷冻防护剂、辐射防护剂、增加渗透性介质,应用非常广泛。胱天蛋白酶(caspase)与真核细胞凋亡密切相关,并参与细胞的生长、分化与凋亡调节,目前仍为研究热点。DMSO对不同细胞caspase有不同的影响,DMSO作为药物或者辅助用药时,可通过抑制caspase对肝细胞有保护作用;对心肌、气道上皮细胞和骨骼肌细胞无毒性,甚至可能通过抑制caspase对心肌和气道上皮细胞还有保护作用;对成骨前体细胞和耳蜗毛细胞依浓度的增加则作用不同;对肿瘤细胞的作用相对复杂;通过促进caspase的表达对白细胞产生损伤作用。在临床和科研中选择DMSO作为药物、诱导剂、有机溶剂和渗透性介质时需考虑DMSO作用于不同器官细胞caspase的复杂影响,从而慎重决定是否选择DMSO。
Dimethyl sulfoxide( DMSO) not only can be used as a drug for its anti-inflammatory and analgesic effect,but is also used widely as inducer,organic solvent,freezing and protective agent,radiation protection agent,increasing permeability medium because of its features. Cysteinyl aspartate specific proteinase( caspase) is closely related to the apoptosis of eukaryotic cells,and participates in cell growth,differentiation and apoptosis regulation,which is still as the research hot spot. DMSO has different effects on the caspase protein of different cells. DMSO has a protective effect as a drug or auxiliary medication on the liver cells by the inhibition on caspase; it is non-toxic to myocardium and airway epithelial cells,and DMSO even has a protective effect on myocardium and airway through the inhibition on caspase protein; DMSO has different effects on osteoblast precursor cells and cochlear hair cells when its concentration increases; the effects on tumor cells is complex; it increases the expression of caspase which damages the white blood cells. So we must take into account the complex influences of DMSO and decide whether to choose DMSO asa drug,inducer,organic solvent and permeable medium in the clinical and scientific research.
