摘要
目的探讨富血小板血浆(platelet-rich plasma,PRP)对于皮肤光老化成纤维细胞分泌功能的影响、初步讨论可能的作用机制。方法适宜浓度PRP处理经中波紫外线(ultraviolet B,UVB)反复照射构建的成纤维细胞光老化模型,24h后在基因和蛋白水平分别检测PRP对于光老化成纤维细胞I型胶原蛋白、基质金属蛋白酶家族、以及细胞内主要抗氧化酶体系的影响。结果 PRP能够上调光老化成纤维细胞的I型胶原蛋白表达水平,抑制由于光老化所引起的基质金属蛋白酶的增高。增加抗氧化酶体系中谷胱甘肽蛋白表达。结论 PRP能够通过增加抗氧化酶体系中谷胱甘肽的表达,改善UVB照射所引起的氧化应激对于皮肤成纤维细胞分泌功能的改变。
Objective To explore the effect o f platelet-rich plasma (PRP) on secretion function of skin photoaging fibroblasts and the mechanism. Methods Fibroblast photo-aging model created by UVB irradiation repeatedly was disposed by PRP of a preference concentration, 24 hours later, the effect of PRP on collagen-I, matrix metalloproteinases (MMPs), and major cellular antioxidase system was detected at gene and protein level respectively. Results PRP up-regulated the exp ression of collagen-I of skin photoaging fibroblasts and inhibited the increase of MMPs caused by UVB irradiation, it increased the expression of protein of glutathione involved in the antioxidant system. Conclusions PRP can improve the changes in secretion function of FBs caused by oxidative stress due to UVB irrational via increasing the expression of protein of glutathione.
