响应面法优化鲟鱼皮胶原蛋白多肽的酶解工艺 被引量:10

Optimization extraction of enzyme soluble collagen peptides from sturgeon by response surface methodology

摘要 为了解决胶原蛋白多肽制备得率低的问题,以鲟鱼-甲骨板皮胶原蛋白多肽得率为主要指标,在单因素试验基础上,进一步利用响应面模型试验确定加酶量、提取时间、液料比3个因素的最优条件,并建立了关系模型。试验结果表明:影响鲟鱼鱼皮胶原蛋白多肽得率的3个因素主次顺序为:液料比>酶解时间>加酶量,最优提取工艺为:0.5 mol/L的乙酸与原料之间的液料比为14.02,加酶量为1.69%,提取时间为8.51 h。在此条件下胶原蛋白多肽得率为78.86%,且产物主要为小分子胶原蛋白,峰值分子量为3719 u。 To solve the problem of low collagen peptides yield, response surface methodology was used, taking the yield of collagen peptides extracted from sturgeon as the main indicators. Based on the single factor experiment, the optimal conditions of enzyme amount, extraction time and liquid ratio were further determined by response surface model, establishing the relationship model at the same time. The results showed that the main factor affecting the collagen peptides yield from sturgeon skin was liquid ratio, followed by enzyme amount and extraction time. The optimal extraction condition was as follows: the ratio between 0.5 mol/L of acetic acid and raw material was 14.02 with 1.69% of enzyme dosage and extracted for 8.51 h, under which condition 78.86% of collagen peptides yield was obtained. Meanwhile, the main products were small molecular collagen and the peak molecular weight was 3719 u.
