BSEP在辅助生殖技术助孕的ICP患者胎盘中的表达及意义

Significance and expression of BSEP in placenta of ICP patients treated with assisted reproductive technology

目的探讨胆盐输出泵(BSEP)在ART助孕和正常受孕妊娠期肝内胆汁淤积症(ICP)患者胎盘组织中的表达及意义。方法采用Western blot和RT-PCR方法检测我院收治的15例自然受孕ICP患者(ICP组)、15例ART助孕ICP患者(ART组)及9例正常妊娠患者(对照组)胎盘组织中BSEP蛋白和m RNA的表达情况,分析三组ICP胎盘中BSEP表达的差异。结果 Western blot和RT-PCR法显示BSEP蛋白和m RNA在自然受孕ICP胎盘中表达分别为(0.443±0.102)和(8.139±4.490),在ART来源胎盘中表达分别为(0.490±0.117)和(9.992±5.253),而正常妊娠胎盘组织中表达明显减少(0.161±0.013)和(1.038±0.271),ICP组和ART组差异无明显差异(P>0.05),而这两组与对照组比较差异有统计学意义(P<0.001)。结论 BSEP蛋白在正常妊娠胎盘组织中微量表达,而在ICP患者胎盘组织中高表达,可能与ICP的发病有关,但自然受孕ICP胎盘和ART助孕ICP胎盘中BSEP蛋白无差异,提示ART助孕并不增加子代妊娠期ICP的发病风险。

Objective To explore the significance and the expression of bile salt export pump（BSEP） in placenta of pa- tients with intrahepatic cholestasis of pregnancy（ICP） of natural conception or pregnancy through ART treatment. Meth- ods The expression of BSEP proteion and mRNA in placenta of 15 ICP patients of natural conception（ICP group）, 15 ICP patients treated with ART（ART group） and 9 normal cases of natural conception（control group） attended our hospital was detected by Western blot and RT-PCR. The difference of BSEP expression in three groups of ICP placenta was analyzed. Results The Western blot and RT-PCR showed that the expression of BSEP protein and mRNA in ICP pla- centa of natural conception was （0.443±0.102） and （8.139±4.490）, respectively. The expression in the placenta of ART group was （0.490±0.117） and （9.992±5.253）, respectively. The expression in the placenta of Control group was significant less, which was （0.161±0.013） and （1.038±0.271）, respectively. There was 21o significant difference between ICP group and ART group（P〉0.05）, but both of them showed statistically significant difference compared to control group（P〈0.001）. Conclusion Trace expression of BSEP protein was found in placenta tissue of normal pregnancy, while high expression in placenta tissue of ICP patients implicated that BSEP protein may related to the occurenee of ICP. There was no dif- ference in expression of BSEP protein in ICP placenta between natural conception and pregnancy with ART treatment,. which suggested that ART will not increase the risk of ICP in offspring.
