摘要
目的:通过系统药理学的方法挖掘影响中药寒热药性的关键靶标,为进一步研究寒热药性的实质提供新的思路。方法:构建寒热中药成分数据库;通过活性成分筛选及网络打靶构建有效成分-靶点网络;通过网络分析筛选出关键靶点,并以此构建靶点-疾病网络。结果:通过网络分析得到寒性中药网络中有PTGS2、PTGS1和SCN5A关键靶点,热性中药网络中有GABRA1、PTGS2和PTGS1关键靶点;寒性中药对于前列腺癌、乳腺癌等癌症及心血管疾病具有潜在的治疗作用,而热性中药在阿兹海默症、疼痛、认知障碍、焦虑症、帕金森症等精神类疾病方面显示出一定的应用价值。结论:基于系统药理学的研究方法有助于寻找寒热药性的关键靶点和疾病网络,为深入诠释寒热药性的生物学本质提供有益的信息和数据支撑。
Objective:Through method of system pharmacology to mine key targets which influence cold-heat nature of TCM,and to provide new ideas for the further study of its essence.Methods:The ingredient database of cold-heat nature of TCM was established.The effective ingredient-target network was further constructed through screening active ingredients and targets.The key targets were selected by network analysis,and the target-disease network was constructed.Results:Through network analysis,PTGS2,PTGS1 and SCN5 A were the key targets in the cold TCM network,and GABRA1,PTGS2 and PTGS1 were the key targets in the heat TCM network.Cold TCM had a potential therapeutic effect on cardiovascular diseases,prostate cancer,breast cancer and other cancers.While heat TCM had shown a certain value of application in Alzheimer's disease,pain,cognitive disorder,anxiety,Parkinson's disease and other mental diseases.Conclusion:The method based on system pharmacology could help to find the key targets of cold-heat nature and disease network of TCM,and provide useful information and data support for giving a further interpretation of the biological essence of cold-heat nature of TCM.
