摘要
目的:研究重楼皂苷Ⅰ对人肺癌NCI-H661细胞凋亡的影响及机制。方法:WST-1法测定重楼皂苷Ⅰ对NCI-H661活性的影响,流式细胞术测定该药对NCI-H661凋亡的影响,Western Blot检测该药对Caspase-8、Caspase-9、Bcl-2蛋白表达的影响,荧光显微镜观察该药对NCI-H661、大鼠皮质海马神经元线粒体形态的影响。结果:重楼皂苷Ⅰ抑制NCI-H661细胞增殖,呈剂量依赖与时间依赖性;该药诱导细胞凋亡,呈剂量依赖性;该药干预后,Caspase-8、Caspase-9、Bcl-2表达有逐渐减少的趋势,以Bcl-2作用最为显著(P<0.05);该药物能快速导致线粒体碎裂。结论:重楼皂苷Ⅰ通过线粒体碎裂诱导NCI-H661凋亡。
Objective:To investigate the effects and mechanisms of Polyphyllin Ⅰ inducing apoptosis of human lung cancer NCI-H661 cells.Methods:Cell viability was detected by WST-1 assay; apoptosis was measured by flow cytometry; the protein expression of Caspase-8,Caspase-9 and Bcl-2 were observed by Western Blot; the morphology of mitochondrial was imaged by fluorescence microscopy.Results:Polyphyllin I significantly inhibited the viability of NCI-H661 cells in the time and dose-dependent manner; the drug significantly and rapidly induced apoptosis in the dose dependence.Western Blot assay showed that the expression of Caspase-8,Caspase-9 and Bcl-2 gradually decreased after exposure to the drug(P〈0.05).We observed that the drug initiated mitochondria fragmentation within short time.Conclusions:Polyphyllin I induces apoptosis of NCI-H661 cells by rapid mitochondrial fragmentation.
