摘要
目的:探讨脾虚痰浊动脉粥样硬化巴马小型猪血清脂蛋白亚类分布的特征并利用PCR Array技术检测主动脉AS信号通路中脂类代谢相关基因表达的变化。方法:9只巴马小型猪随机分为正常组、模型组和治疗组,每组3只,正常组普通饲料喂养,模型组与治疗组给予高热量高脂适量胆盐饲料喂养并进行冠脉球囊挤压伴拉伤及跑步措施进行干预至24周,制备巴马小型猪病症结合脾虚痰浊动脉粥样硬化模型,治疗组除上述造模措施外给予健脾祛痰化瘀方喂饲24周。全自动生化分析仪检测血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C);Lipoprint脂蛋白分类仪对HDL、LDL颗粒进行分类,分析不同大小HDL、LDL颗粒在脾虚痰浊动脉粥样硬化巴马小型猪中的分布特征;PCR Array技术检测脂类代谢相关基因表达。结果:与正常组比较,模型组小型猪血清TG、LDL-C水平显著升高(P<0.05,P<0.01);HDL-C水平显著降低(P<0.05);LDL平均颗粒较小(P<0.01),sd-LDL浓度所占百分比显著升高(P<0.05);大颗粒HDL-C含量显著降低、大颗粒HDL-C占HDL-C浓度百分比显著降低、小颗粒HDL-C含量显著升高,小颗粒HDL-C浓度占HDL-C浓度百分比显著升高(P<0.05,P<0.01)。与模型组比较,治疗组小型猪LDL-C水平显著降低(P<0.05);大颗粒HDL-C含量显著升高、大颗粒HDL-C百分比显著升高、小颗粒HDL-C含量显著降低,小颗粒HDL-C浓度占HDL-C浓度百分比显著降低(P<0.05,P<0.01)。PCR Array技术分析发现模型组小型猪上调≥2倍的基因包括ABCA1、IL4、PTGS1、NR1H3、PPARD,下调<0.5的基因有LDLR、FABP3、LPL、PPARG。结论:LDL和HDL亚类分布异常可能是导致动脉粥样硬化发生的重要原因之一。
Objective:To explore the distribution characteristics of serum lipoprotein subclasses on Bama mini pigs with spleen deficiency and phlegm turbidity atherosclerosis,and to detect the change of related gene expression of lipid metabolism in the aorta AS signal pathway using PCR array.Methods:Nine Bama mini pigs were randomly divided into normal group,model group and treatment group,3 pigs in each group.Normal group was fed with normal diet.Model group and treatment group were fed with high-calorie and high-fat diet,and also were intervened lasting for 24 weeks with the measures of coronary balloon compression with strain and running,for preparing Bama mini pigs atherosclerosis combined with spleen deficiency and phlegm turbidity syndrome model.In addition to the above modeling measures,the treatment groups were fed with Jianpi Qutan Huayu Formula continued to 24 th week.Automatic biochemical analyzer was used to detect TC,TG,LDL-C and HDL-C; Lipoprint lipoprotein classifier was used to classify HDL and LDL particles to analyze the contribution characteristics of HDL and LDL particles in Bama mini pigs with spleen deficiency and phlegm turbidity atherosclerosis; PCR arraywas used to detect related gene expression of lipid metabolism.Results:Compared with normal group,the levels of TG and LDL-C in serum in model group were significantly increased(P〈0.05,P〈0.01),while the level of HDL-C was significantly decreased(P〈0.05).The average particle size of LDL was smaller(P〈0.01),and the percentage of sd-LDL was significantly increased(P〈0.05).The content of large particle HDL-C was significantly decreased; the concentration percentage of large particle HDL-C in total HDL-C was significantly decreased,while the content of small particle HDL-C was significantly increased,the concentration percentage of small particle HDL-C in total HDL-C was significantly increased(P〈0.05,P〈0.01).Compared with model group,the level of LDL-C in treatment group was significantly decreased(P〈0.05).The content of large particle HDL-C was significantly increased,The contentration percentage of large particle HDL-C in total HDL-C was significantly increased,the content of small particle HDL-C was significantly decreased,while the concentration percentage of small particle HDL-C in total HDL-C was significantly decreased(P〈0.05,P〈0.01).PCR Array found the up-regulation genes of mini pigs in the model group which were increased more than 2 times were ABCA1,IL4,PTGS1,NR1 H3,PPARD.While the down-regulation genes were cut less than half,such as LDLR,FABP3,LPL,PPARG.Conclusion:The abnormal distribution of LDL and HDL subclasses might be one of the important reasons for the occurrence of atherosclerosis.
