桑芪首乌方对SHR大鼠脑血管保护作用研究 被引量:2

摘要 目的:探讨中药制剂桑芪首乌方(SQSW)对实验性原发性高血压大鼠血压及脑血管的影响。方法:将10只SD大鼠作为正常组,50只原发性高血压大鼠(SHR大鼠)随机分为高血压组和SQSW组(1.2、2.4、4.8g生药/m L),马来酸依那普利片组作为对照组,每组10只,连续灌胃5周,观察给药后第1、2、3、4、5周SQSW对SHR大鼠平均动脉压(MAP)、舒张期动脉压(DAP)的影响;给药后第5周评分各组大鼠易激惹程度,心脏取血,分离血清,测定乳酸脱氢酶(LDH)、羟肌酸激酶(CK)和超氧化物歧化酶(SOD)及丙二醛(MDA)的值;HE染色观察脑血管壁的变化。免疫组化法检测各组大鼠脑组织切片血管细胞黏附分子(VCAM-1)的表达。结果:SQSW高剂量组大鼠MAP、DAP值显著降低(P<0.05);第5周时SQSW高剂量组大鼠的激惹程度显著低于SHR组(P<0.05);SQSW高剂量组能显著降低血清LDH、CK及MDA值(P<0.05),提高SOD活性(P<0.05)。HE染色显示SQSW能减轻脑血管的玻璃样变,免疫组化结果表明SQSW能明显抑制脑组织中VCAM-1的表达,与SHR组比较差异有统计学意义(P<0.05,P<0.01)。结论:SQSW能够明显降低SHR大鼠血压的MAP、DAP值,改善大鼠的宏观表征,其机制可能与SQSW的抗氧化损伤及减轻血管损伤作用有关。 Objective:To investigate the therapeutic effect of Sangqi Shouwu Decoction(SQSW) on hypertension rats and its possible mechanisms.Methods:Ten SD rats were treated as normal group.Fifty spontaneously hypertensive rats were randomly divided into five groups:SHR group,SQSW(1.2,2.4,4.8 g crude drug/m L) and Enalapril Maleate tablets group.All the groups were givenintragastric administration for 5 weeks continuously.Blood pressure(BP) were measured respectively by tail-cuff technique before and in the 1 st week,2 nd week,3 rd week,4 th week and 5 th week during the therapy.The serum content of LDH,CK,SOD and MDA were detected after 5 weeks.The expression of VCAM-1 were measured by immunohistochemical method.Vascular lesions were detected by HE stain.Results:The MAP value and DAP value of high dose SQSW group decreased significantly(P〈0.05).At week 5,the score of irritability in high dose SQSW group was significantly lower than that in SHR group(P〈0.05).The serum LDH,CK and MDA values in high dose SQSW group were significantly decreased(P〈0.05) and SOD activity was increased(P〈0.05).HE staining showed SQSW could reduce the vitreous changes of cerebral vessels,and immunohistochemistry showed SQSW could significantly inhibit VCAM-1 expression in brain tissues,which was significantly different from SHR group(P〈0.05,P〈0.01).Conclusion:SQSW could significantly reduce blood pressure and improve the macro characterization of rats.Some of the mechanisms might be related to the antioxidant damage of SQSW and the reduction of vascular injury.
