摘要
为了揭示杜仲叶中对结肠癌细胞增殖与凋亡的有效成分,首先对杜仲叶提取物进行了成分分析,然后取其中两种主要成分京尼平苷酸与绿原酸分别对结肠癌细胞株HCT116与LOVO细胞进行了增殖与凋亡的影响实验。结果显示:当绿原酸浓度达1.610 6μg/L时,作用于LOVO细胞的抑制率达94.96%,超过阳性对照药顺铂的抑制效果。而京尼平苷酸抑制效果不显著。绿原酸作用于HCT-116和LOVO细胞的凋亡率分别为24.08%,33.80%,均高于空白对照组的5.83%,13.71%。实验结果证实杜仲叶提取物中绿原酸对结肠癌细胞的增殖与凋亡都有显著影响,是杜仲叶中抑制结肠癌细胞增殖、促进其凋亡的主要成分之一。
To reveal effective components in Eucommia Ulmoides leaves on the proliferation and apoptosis of colon cancer cells. Firstly, the contents of main constituents of Eucommia Ulmoides leaves extract were determinated. The effects on colon cancer cell line HCT116 and LOVO cell proliferation and apoptosis under geniposidic acid and chlorogen- ic acid were observed. Under 1. 6106 g/L of chlorogenic acid, the inhibitory rate on LOVO cell was 94.96%, which was greater than that of cis -platinum (positive control). Corresponding to geniposidic acid had no inhibitory effect. The ap- optosis rates of chlorogenic acid on HCT - 116 and LOVO cells were 24.08% and 33.80%, which were higher than those in blank control group(5.83% and 13.71% ), respectively. From the results analysis, chlorogenic acid induced colon cancer cell apoptosis was significantly higher than that of the blank group, close to the positive control group. All these results indicated that chlorogenic acid is one of the main components in Eucommia Ulmoides leaves to inhibit prolif- eration and promote apoptosis for colon cancer cells.
