参龙煎剂干预肺纤维化大鼠血清IFN-γ、IL-4表达水平影响的实验研究 被引量:4

摘要 目的:观察中药复方参龙煎剂对肺纤维化大鼠血清中IFN-γ、IL-4表达水平的影响,探讨中药复方参龙煎剂对肺纤维化大鼠的疗效及其作用机制。方法:120只Wistar大鼠,随机分为6组,即空白对照组、模型对照组、西药组、中药高剂量组、中药中剂量组、中药低剂量组。西药组予以醋酸泼尼松龙片0.625 mg,中药高、中、低剂量组分别予以1.0、0.5、0.25 g剂量,按照1 m L/100 g体积灌胃。每组中分别于造模次日起第7、14、28天3个时间点随机选取6只,进行取材。通过HE染色观察病理组织变化和肺纤维化分级情况,ELISA法测定血清中IFN-γ、IL-4含量的变化。结果:与空白对照组相比,模型对照组、中药高剂量组和中药低剂量组大鼠血清的IL-4在3个时间段均明显升高(P〈0.01或P〈0.05);与模型对照组相比,西药组、中药高剂量组、中药中剂量组、中药低剂量组大鼠血清的IL-4在3个时间段均明显降低(P〈0.01);与空白对照组比较,其余各组大鼠肺组织IFN-γ在3个时间段表达水平显著下调(P〈0.01或P〈0.05);与模型对照组比较,西药组、中药高剂量、中剂量组大鼠肺组织IFN-γ在3个时间段表达水平显著上调(P〈0.01)。结论:中药复方参龙煎剂可通过抑制IL-4、增强INF-γ表达以达到抗肺纤维化的目的,其内在机制可能与调节Th1/Th2细胞因子网络失衡有关。 Objective : To observe the effect of Compound Shenlong Jianji on expressions of IFN -γand IL - 4 in pul- monary fibrosis rats and investigate the effect of Shenlong Jianji on pulmonary fibrosis in rats and its mechanism. Method: Totally 120 Wistar rats were randomly divided into 6 groups : blank control group, model control group, western medicine group, high dose group, middle dose group and low dose group. The western medicine group used Prednisolone Acetate Tablets O. 625 mg. Chinese high, medium and low dose groups were treated with 1.0 g, 0.5 g, 0.25 g of Compound Shenlong Jianji, according to l mL/100g volume by gastric lavage. In each group we detected at the 7d, 14d, 28d three time points randomly and selected 6 rats. HE staining was used to observe the changes of pathological tissue and pulmo- nary fibrosis, and the changes of IFN - -y and IL - 4 in serum were measured by ELISA. Result : Compared with the blank control group, the serum levels of IL - 4 in the model control group, the high dose group and the low dose group were sig- nificantly higher than those in the three time points (P 〈 0. 01 or P 〈 0. 05). Compared with the model control group, the serum IL -4 in the western medicine group, the high dose group, the middle dose group and the low dose group were sig- nificantly decreased in the three time points(P 〈0.01 ). Compared with the blank control group, the expression level of IFN - X lung tissue of the rats in the other groups was significantly lower in the three time points( P 〈 0. 01 or P 〈 0. 05 ). Compared with the model control group, the expression level of IFN - γ lung tissue of rats in western medicine group, high dose and middle dose group was significantly increased in three time points( P 〈 0. 01 ). Conclusion:Shenlong Jianji can inhibit IL - 4 and enhance IFN - γ expression in order to achieve the purpose of anti pulmonary fibrosis. The mecha- nism might be related to the regulation of Thl/Th2 cytokine network imbalance.
