沉默Cx43基因对左归丸促MC3T3-E1细胞分化作用的影响

Effect of Cx43 Gene Silencing on Promoting Differentiation of MC3T3-E1 Cells Induced by Zuogui Pill

目的：研究沉默Cx43基因对左归丸含药血清促MC3T3-E1细胞分化作用的影响。方法：采用慢病毒介导的RNA干扰技术沉默Cx43基因,将培养细胞分为正常对照A组、含药血清B组、基因沉默C组。细胞培养第3、5、7、9天进行ALP活性测定、第21天矿化结节茜素红染色,进行组间对比。结果：左归丸含药血清可以促进MC3T3-E1细胞的分化,与A组比较：ALP活性增强,其中第5、7、9天差异显著（P〈0.05）;茜素红染色面积增加（P〈0.01）。C组与B组比较：ALP活性降低,其中第7、9天差异显著（P〈0.05）;茜素红染色面积明显减少（P〈0.05）。结论：沉默细胞Cx43基因导致左归丸含药血清促MC3T3-E1细胞分化作用明显下降。

Objective： To study the effect of Cx43 gene silencing on the promoting differentiation of MC3T3 -E1 cells induced by Zuogui Pill medicated serum. Methods ： First, lentivirus - mediated RNA interference technique was used to silence the Cx43 gene and then the cultured cells were divided into normal control group A, serum group B and gene silen- cing group C. ALP activity was measured and compared at 3,5,7 and 9 days, and mineralized nodules by alizarin red stai- ning were detected and compared at day 21. Results： Zuogui Pill medicated serum can promote the differentiation of MC3T3 - E1 cells. Compared with A group, ALP activity increased, of which the 5,7 and 9 days were significantly differ- ent （P 〈0. 05） ;and the alizarin red staining area increased（P 〈0. 01 ）. C group compared with B group,ALP activity decreased, and at 7 and 9 day were significantly different （P 〈 0. 05 ） ;and the alizarin red staining area was significantly reduced （P 〈0. 05 ）. Conclusion： Zuogui Pill medicated serum on MC3T3 - E1 cell promoting differentiation was signifi- cantly decreased by silencing of Cx43 gene.
