绞股蓝总苷对氧化低密度脂蛋白损伤血管内皮细胞的保护作用研究 被引量:6

摘要 目的:探讨绞股蓝总苷对氧化低密度脂蛋白损伤血管内皮细胞的保护作用的研究。方法:采用ox-LDL对血管内皮细胞造成损伤模型,并将HUVEC分为空白对照组、模型组、绞股蓝高、中、低剂量组,空白对照组给予常规DMEM液+5%血清,模型组除与空白对照组相同常规DMEM液+5%血清外,予加入100mg/L ox-LDL干预,绞股蓝低、中、高剂量组在分别给予10、50、100 mg/L绞股蓝提取物24 h后,加入100mg/L ox-LDL。观察各组细胞生存活性、线粒体膜电位、iNOS、MMP-9、NF-κB p65、NO、SOD水平等指标变化,其结果应用统计学软件SPSS 19.0进行数据分析。结果:与空白对照组相比,模型组细胞生存活性、线粒体膜电位、NO、SOD等指标水平明显低于空白对照组,iNOS、MMP-9、NF-κB p65等指标水平明显高于空白对照组;对模型组相比,绞股蓝低、中、高剂量组细胞生存活性、线粒体膜电位、NO、SOD等指标水平明显高于模型组,iNOS、MMP-9、NF-κB p65等明显低于模型组。以上结果差异均有统计学意义(P〈0.05)。结论:绞股蓝总苷具有保护血管内皮细胞活性,维持线粒体膜电位水平,提高血管内皮细胞抗氧化活性等药理作用,并通过以上作用机制起到对氧化低密度脂蛋白损伤的血管内皮细胞的保护作用。 Objective : To explore the protection effect of gypenosides of oxidized on vascular endothelial cell damage induced by low density lipoprotein. Methods : The ox - LDL was used to set up vascular endothelial cell damage model and HUVECs were divided into blank control group, model group, and gypenosides of high, medium and low dose groups. Blank control group was given conventional liquid DMEM + 5% serum. Model group in addition to the same as the b|ank control group' s routine DMEM liquid + 5% serum, added 100 mg/L ox - LDL intervention. The gypenosides low, medi- um and high dose groups were respectively given 10, 50, 100 mg/L gypenosides extract after 24 h and added 100 mg/L ox - LDL. We observed each group' s cell survival activity and mitochondrial membrane potential, iNOS, MMP - 9, the NF- KB p65, NO, SOD level indicators changes and the result was analyzed by statistical software SPSS 19.0. Results: Compared with the blank control group, the model group' s cell survival activity, mitochondrial membrane potential, NO and SOD levels were significantly lower than those of the blank control group. The iNOS, MMP - 9 and the NF - KB p65 index levels were significantly higher than those of the blank control group. Compared with the model group, gypenosides low, medium and high dose groups' cell survival, mitochondrial membrane potential, NO and SOD activity index level were significantly higher than those of model group ; iNOS, MMP - 9 and the NF - KB p65 were significantly lower than those of the model group, etc. The above results difference hade statistical significance(P 〈0. 05 ). Conclusions: Gype- nosides have protect effect on vascular endothelial cell activity, maintain a level of mitochondrial membrane potential and improve the antioxidant activity pharmacological effects, such as vascular endothelial cells and through the above mecha- nism has the oxidation of low density lipoprotein of vascular endothelial cells injury protection.
