铜绿假单胞菌的血清分型、鞭毛分型和脉冲场凝胶电泳分型 被引量:6

Serotyping,flagella typing and pulse field gel electrophoresis of Pseudomonas aeruginosa isolates

摘要 铜绿假单胞菌(Pseudomonas aeruginosa,PA)是一种重要的人畜共患病原菌,给人畜健康带来威胁。细菌分型是揭示病原菌流行规律、传播机制以及制定相应防控措施的基础。选取水貂、人和羊源PA共78株分别进行O-抗原血清分型、H-抗原鞭毛分型以及脉冲场凝胶电泳(PFGE)分型。结果显示,60株水貂源PA主要流行血清型为G型(73.3%),其次为I,M,B,E型;鞭毛A型和B型分别占91.7%和8.3%。16株人源PA以血清型E型(25%)为主,其他型别G,M,I,B,D,F,H零散分布,鞭毛A,B型分别占68.8%和31.2%。2株羊源PA血清型为I型(100%),且均为鞭毛B型。PFGE分子分型共分为14个谱型,24个亚型,其中B1、F3、G1、H1这4个亚型的菌株同源性达到100%。结果表明,水貂源PA感染与血清G型和鞭毛A型高度相关(P〈0.01);人源PA感染优势血清型为E型且与鞭毛A型显著相关(P〈0.01);2株羊源PA感染均为血清I型和鞭毛B型;不同来源的PA菌株其PF—GE分型呈现多样化和复杂性,相同来源的菌株其PFGE谱型有的完全相同,显示同一个克隆传播的可能,有的差异很大,显示不同分子克隆传播。本试验有助于阐明山东省3种来源PA的分子流行特征,为开展PA相关风险评估、控制及疾病诊治提供依据。 Pseudomonas aeruginosa (PA) is one of the important zoonotic pathogens that commonly cause a great threat to human and animal health. Bacterial typing is the basis for revealing the epi- demiological rules, the mechanisms of disease transmission and developing appropriate control measures. Totally 78 PA isolates originated from mink, human and sheep were analyzed by O-anti- gen serotyping, H-antigen flagella typing, and pulse field gel electrophoresis. The results showed that the predominant serotype of 60 isolates from mink was type G (73.3%) ,followed by I,M,B and E. Besides,91.7% and 8.3% of these isolates were found to be flagella A and B,respectively. The main serotype of 16 PA isolates originated from human was type E(25%) and other serotypes (G,M,I,B,D,F and H) had scattered distribution with 68.8% flagella A and 31.2% flagella B, respectively. Two isolates originated from sheep were both serotype I and flagella B. There were totally 14 types, 24 subtypes from selected 48 isolates by PFGE analysis, and isolates from B1 ,F3 ,G1 and H1 were of the same PFGE bands with homology up to 100%. Our results demon- strated that PA infection of mink was highly correlated with isolates of serotype G and flagella A (P〈0.01) ,while PA infection of human was closely related with isolates of serotype E and flagel- la A(P〈0.01). Two isolates of sheep was closely related with serotype I and flagella B. The PF- GE typing showed the characteristic with complexity and diversity among strains from different sources. Some isolates from the same sources showed 100% similarity and exactly the identical PF- GE band,indicating the possibility of one same clone spreading,while some isolates vary greatly, showing different molecular clonal transmission. Our study will be helpful to determine the molec- ular epidemiology of PA from three species in Shandong province, and provide certain laboratory data for risk assessment,control and prevention of PA infections.
