T细胞亚群在特发性炎性肌病动物模型骨骼肌中的表达

摘要 目的分析T细胞亚群在实验性自身免疫性肌炎(EAM)动物模型骨骼肌组织中的表达情况,研究T细胞亚群在特发性炎性肌病(IIM)发病机制中的作用和意义。方法用异种骨骼肌匀浆免疫诱导的方法建立EAM大鼠模型,10例健康大鼠作为正常对照,采用连续切片免疫组织化学染色方法,计算机图像分析检测CD_3、CD_4、CD_8在EAM动物模型组和正常对照组骨骼肌组织中的表达及其变化情况。结果 CD_3、CD_4、CD_8在肌炎模型的横纹肌、血管周围的炎细胞表达,两组间差异具有显著性(P<0.05)。结论 EAM模型制备成功,提示CD_3、CD_4、CD_8可能参与了EIM骨骼肌损伤的发生和发展。 Objective: To study the expression of T cell subgroup in muscle tissues of experimental autoimmune myositis (EAM) model in rats. To investigate the role and significance ofT cell subsets in the pathogenesis of idiopathic inflammatory myopathy (IIM) Methods:Immunizing with homogenate from rabbit skeletal muscles induced EAMs.Tissue specimens of limbs skeletal muscles were taken from 30 SD rats of EAM and 10 healthy rats as control. Immunohistochemical staining using a set of polyclonal antibodies against CD3.CD4.CD8 identified the expression of them in these tissues, Results:In the capillary, vein wall, arterial endothelial cell and nerve sheath membrane of the EAM group, CD3.CD4.CD8 was detected, The animal model was significantly higher than those from control (p〈0,05) .Conclusion:The establishment of EAM animal model is successful. CD3. CD4.CD8 may play an important role in the pathogenesis of EAM.
