摘要
目的研究桂枝茯苓对子宫内膜异位症(EMs)大鼠异位内膜组织生存素(Survivin)和半胱氨酸蛋白-3(Caspase-3)表达的影响。方法自体移植法建立EMs模型,随机分为模型组、对照组和低、中、高剂量实验组,每组15只。模型组均予以0.9%NaCl 2 mL;对照组予以0.52 mg·mL^(-1)米非司酮混悬液2 mL;低、中、高剂量实验组分别予以29,58和116 mg·mL^(-1)桂枝茯苓混悬液2 mL。5组大鼠均予以灌胃给药,每天1次,连续用药28 d。取子宫内膜组织,用免疫组织化学法检测Survivin和Caspase-3的表达量。结果模型组、对照组和低、中、高剂量实验组的Survivin阳性表达率分别为92.86%,26.67%,60.00%,33.33%和28.57%,Caspase-3阳性表达分别为14.29%,86.67%,46.67%,73.33%和85.71%,对照组和低、中、高剂量组的上述指标与模型组比较,差异均有统计学意义(均P<0.05)。结论桂枝茯苓治疗EMs的作用机制可能是通过下调Survivin在EMs中的高表达,激活细胞中的Caspase-3活化途径,从而抑制异位内膜的增长。
Objective To study the effect of Guizhi Fuling on the expression of Survivin and Caspase-3 in endometriosis( EMs) rats.Methods The EMs model was built by autologous transplantation. The rats were randomly divided into model group,control group,low-dose,middle-dose and high-dose test groups,with 15 rats per group. Model group was treated with 0. 9% NaCl 2 mL. Control group was given 0. 52 mg·mL^(-1) mifepristone suspension 2 mL. Low-dose,middle-dose and high-dose test groups were treated with 29,58 and 116 mg·mL^(-1) Guizhi Fuling suspension 2 mL,respectively. Five groups rats were given drugs by intragastric administration,qd for 28 days. The expressions of Survivin and Caspase-3 were detected by immunohistochemistry.Results In the model group,control group,low-dose,middle-dose and high-dose test groups,the positive expression rates of Survivin were92. 86%,26. 67%, 60. 00%, 33. 33% and 28. 57%, the positive expression rates of Caspase-3 were 14. 29%,86. 67%,46. 67%,73. 33% and 85. 71%. Compared with model group,the above indicators in the control group and low,medium and high dose groups had statistically significant differences( all P 0. 05). Conclusion Themechanism of Guizhi Fuling in the treatment of EMs may be the high expression of Survivin and the activation of cells in Caspase-3 activation pathway,which induces ectopic endometrial cells atrophy,necrosis and apoptosis,thus inhibiting the growth of ectopic endometrium.
