摘要
目的:探讨ENAH对肝癌细胞侵袭和迁移能力的影响及其机制。方法:用实时定量聚合酶链反应(RT-PCR)的方法在mRNA水平测定ENAH在48对结肠癌组织及正常组织中的表达;改变ENAH表达后检测肝癌细胞系中ENAH,Transwell实验检测其侵袭和迁移能力。结果:RT-PCR结果显示:ENAH在肝癌组织中的表达高于正常组织;相比正常肝细胞系,ENAH在Huh7及BEL7402中的表达升高。下调ENAH在Huh7及BEL7402中的表达后,Transwell实验结果显示肝癌细胞系Huh7及BEL7402的运动、侵袭和迁移能力均明显减弱。结论:ENAH能够促进肝癌细胞系Huh7和BEL7402的侵袭和迁移能力。
ObjectiveTo investigate the influence of ENAH on the invasion and migration in human liver cancer cell lines. Methods: The mRNA levels of ENAH in 48 pairs of the liver cancer tissues and adjacent normal tissues, BEL7402 and Huh7 were determined by real - time PCR( RT - PCR) assays. The influence of ENAH on the mobili-ty, invasion and migration of liver cancer cell lines were detected using Transwell assay. Results: The results of RT - PCR showed that ENAH were up - regulated in liver cancer tissues compared with adjacent normal tissues. The ex-pression level of ENAH in Huh7 was significantly higher than that in THLE3, Transwell assay showed that the mobile, invasive and migratory ability were decreased after down - regulated the expression of ENAH in the liver cancer cell line Huh7. Conclusion : ENAH could enhance the invasive and migratory ability of liver cancer cell Huh7.
