摘要
目的:探讨透骨消痛胶囊对膝骨关节炎大鼠关节软骨G蛋白偶联信号传导系统的影响。方法:2月龄清洁级雄性SD大鼠36只,喂养1周后,随机分为空白组12只和造模组24只。造模组在第1,4,7天双膝关节腔注射0.2 mL质量分数为4%的木瓜蛋白酶。造模成功后,抽签法随机分为模型组和透骨消痛胶囊组,每组12只。透骨消痛胶囊组按照3.6 g·kg^(-1)·d^(-1)的剂量灌服透骨消痛胶囊,空白组、模型组给予等量生理盐水。12周后取关节软骨,采用Western blot技术检测关节软骨中Gαs、Gα(i)、Gα(pan)蛋白表达。结果:与空白组比较,模型组Gαs、Gα(pan)表达量下降,Gα(i)表达量上升,差异有统计学意义(P<0.01);与模型组比较,透骨消痛胶囊组Gαs、Gα(pan)表达量上升,Gα(i)表达量下降,差异有统计学意义(P<0.01或P<0.05)。结论:透骨消痛胶囊可能通过激活G蛋白偶联信号传导系统,延缓关节软骨的退变。
Objective:To investigate the effect of Tougu Xiaotong Jiaonang(透骨消痛胶囊)on G protein coupled signal transduction system of articular cartilage in rats with knee osteoarthritis.Methods:Thirtysix two-month male SD rats fed for a week were randomly divided into a blank group(12 rats)and a model group(24 rats).The model group was injected with 0.2 mL of papain(with mass fraction of 4%)into articular cavities of both knees at the first,fourth,and seventh days.After the the models were established,they were randomly divided into a model group and a Tougu Xiaotong Jiaonang group,twelve rats in each group.The Tougu Xiaotong Jiaonang group was drenched with Tougu Xiaotong Jiaonang(3.6 g·kg^(-1)·d^(-1)),while the control group and the model group were given normal saline.After twelve weeks,the articular cartilage was taken and the expressions of protein Gαs,Gα(i)and Gα(pan)in articular cartilage were detected by Western blot technique.Results:Compared with the control group,the expression of Gαs and Gα(pan)in the model group decreased,while the expression of Gα(i)increased,and the difference was statistically significant(P < 0.01).Compared with the model group,the expressions of Gαs and Gα(pan)in the Tougu Xiaotong Jiaonanggroup increased and the expression of Gα(i)decreased,and the difference was statistically significant(P<0.01 or P<0.05).Conclusion:Tougu Xiaotong Jiaonang may delay the degeneration of articular cartilage by activating G protein coupled signal transduction system.
