摘要
普那霉素是一种新型的抗生素,对大多数革兰氏阳性菌和部分革兰氏阴性菌有强烈的抑制作用,是万古霉素的替代药物。对Streptomyces pristinaespiralis LS15产普那霉素的发酵培养基进行了PB-CCD的优化研究。PB试验表明,K_2HPO_4、酵母抽提物和Mg SO_4是对摇瓶发酵普那霉素具有显著影响的3个因素。然后进行的爬坡试验和中心组合试验表明,当K2HPO4质量浓度2.34 g/L,酵母抽提物质量浓度6.52 g/L,MgSO_4质量浓度1.84 g/L时,普那霉素的发酵产量最高达到2.23 g/L,比起初始培养基提高了86.4%。对Streptomyces pristinaespiralis LS15代谢过程中的酶活测定表明,HK,PK,AK,CS等在优化培养基中的酶活均高于初始培养基,是普那霉素产量提高的原因之一。
Pristinamycin is a new kind of antibiotics, which can inhibit majority of gram-positive bacteria and some gram-neg-ative bacteria,being considered as the replacement of vancomycin. In this paper, the PB-CCD research was employed to op-timize fermentation medium for Streptomyces pristinaespiralis LS15 producing pristinamycin. PB experiment showed that K2 HPO4,yeast extract and MgS O 4 were significant factors for shake flask fermentation. Then the climbing experiment and CCDshowed that when concentration of K2 HPO4 2.34 g/L,yeast extract 6.52 g/L, MgS O 4 1.84 g/L, the yield of pristinamycin reached 2.23 g/L, which increased by 86.4% compared to initial medium. Determination of enzyme activity in Streptomyces pristinaespiralis LS15 showed that HK, PK, AK, CS, etc in the optimized culture medium were higher than those in initial medium, which was one reason for the increased production.
