摘要
活性氧(ROS)是植物有氧代谢的副产物,本身具有毒性。H2O2作为ROS家族中的一员,在植物抵御病虫害侵染过程中扮演着重要角色。为了探明活性氧途径相关基因在高粱对高粱蚜的防御反应中所发挥的作用,以高粱抗蚜品种HN 16(包含RMES1基因)和感蚜突变体(RMES1突变)作为研究材料,对蚜虫取食过程中(0,24,48,72h和96h)抗坏血酸过氧化物酶基因(Sb 002G431100)的表达模式进行了分析。结果表明,在蚜虫取食的最初24h内,该基因在HN 16和感蚜突变体间没有明显差异,但当蚜虫取食48h后,HN 16中的表达量显著高于突变体,蚜虫取食72h后突变体中的表达量显著下降,但在HN 16中则始终维持在相对稳定的水平,说明该基因在RMES1介导的抗虫反应中发挥着积极的作用。
Reactive oxygen species (ROS) are byproducts of plant aerobic metabolism and are toxic in themselves.As a member of the ROS family, H2 02 plays an important role in the process of plant pro- tection against diseases and insect infestation. In order to find out the role of related genes in ROS pathway in sorghum defense response to Sorghum Aphid's, HN 16 (carrying RMES1) and the mutant susceptible to M. sacchari were used to study the expression pattern of ascorbate peroxidase gene (Sb 002 G 431100) in the aphid feeding process (0,24,48,72 h and 96 h).The results showed that there is no significant difference in the gene expression between HN 16 and the mutant within the first 24 h aphids feeding,but after aphids feeding 48 h,the gene expression in HN 16 was significantly high- er than that of mutant.The expression in mutant significantly decreased after aphid feeding 72 h,but that maintained at a relatively stable level in HN 16.That means that the gene plays a positive role in the resistance reaction to aphid mediated by RMES1.
