人参皂苷Rh1与Rg1对脂肪细胞增殖分化的影响比较

摘要 目的比较人参皂苷Rh1与Rg1对3T3-L1细胞增殖分化的影响,为进一步研究Rh1对糖脂代谢的影响奠定基础。方法以不同浓度的Rh1与Rg1(25、50、100μmol/L)处理3T3-L1细胞,以MTT比色法检测两者对细胞生长的影响。鸡尾酒法诱导3T3-L1细胞向成熟的脂肪细胞分化,诱导开始时即加入上述浓度的Rh1与Rg1,细胞分化成熟后测定细胞成脂程度,并测定细胞内PPARγ及脂联素基因和蛋白表达的变化。结果 (1)Rg1和Rh1均促进3T3-L1细胞的增殖,且在中高浓度下的Rh1更能促进细胞生长。(2)两者均抑制3T3-L1细胞的成脂分化,且Rh1的抑制作用更强。(3)与Rg1相比,Rh1在中高浓度下下调PPARγm RNA以及上调脂联素m RNA的作用都更强。(4)Rh1和Rg1均减少PPARγ的蛋白表达,增加脂联素的蛋白表达,且在中高浓度下Rh1的作用更显著。结论人参皂苷Rh1对3T3-L1细胞的促增殖作用以及抑制分化作用都强于Rg1,且Rh1能显著下调细胞内PPARγ基因和蛋白的表达,并促进脂联素基因和蛋白的表达。 Objec t ive To compare ginsenoside Rh1 with ginsenoside Rg1 in the effect on the proliferation and differentiation of 3T3-L1 cells and to lay a foundation for further study on the effect of Rh1 on glucolipid metabolism. MeAods 3T3-L1 cells were treated with Rh1 and Rg1 different concentrations of 25 μ mol/L, 50 μ mol/L and 100 μ mol/L, and MTT colorimetric method was applied to measuring the effects of Rh1 and Rg1 on the growth of cells. The cocktail method was used to induce the 3T3-L1 cells to differentiate into adipocytes. At the beginning of induction, Rh1 and Rg1 in above concentrations were separately added into the cells; when the cells maturated, the adipogenic degree was measured, and the changes in the mRNA and protein expressions of peroxisome proliferator- activated receptor gamma (PPAR7) as well as the adiponectin in cells were measured. Results ⑴ Both Rg1 and Rh1 promoted the proliferation of 3T3-L1 cells and Rh1 in moderate and high concentrations could better promote the growth of cells.(2) Both Rg1 and Rh1 inhibited the adipogenic differentiation of 3T3-L1cells, while Rh1 had better inhibitory effect. (3) Compared with Rg1, Rh1 in moderate and high concentrations had better effects in reducing PPAR mRNA and increasing adiponectin mRNA.(4) Both Rh1 and Rg1 reduced the protein expression of PPAR and increased the protein expression of adiponectin, while Rh1 in moderate and high concentrations had better effect. Con clusion The ginsenoside Rh1 has better effect than ginsenoside Rg1 in promoting the proliferation and inhibiting the differentiation of 3T3-L1 cells, and can significantly reduce the mRNA and protein expressions of PPAR γ in cells while promoting the mRNA and protein expressions of adiponectin.
