UPLC法测定20种滴眼液中的9种常用抑菌剂 被引量:7

摘要 建立了超高效液相色谱法测定滴眼液中的9种常用抑菌剂。采用Hypersil GOLD色谱柱,以0.5%三乙胺溶液(用磷酸调至p H 3.0)∶甲醇为流动相,线性梯度洗脱,使用UPLC-PDA检测,检测波长260 nm,结合保留时间和紫外光谱定性。结果 9种抑菌剂可完全分离,回收率为98%~102%,RSD为0.1%~0.4%。在20种滴眼液中检出苯氧乙醇、苯扎溴铵、对羟基苯甲酸乙酯和苯扎氯铵4种抑菌剂。该方法准确、快速,灵敏度高,可作为滴眼液中常用抑菌剂的控制方法。 A UPLC method was established for the determination of 9 commonly used bacteriostatic agents in eye drops. A Thermo Hypersil GOLD column was used, with the mobile phase of 0.5% triethylamine (adjusted to pH 3.0 with phosphoric acid)∶methanol with linear gradient elution, at the detection wavelength of 260 nm. The qualitative analysis was carried out based on retention time and spectrum by UPLC-PDA. Under the above conditions, nine bacteriostatic agents were separated completely. Their recoveries were 98%—102%, with RSDs of 0.1%—0.4%. Four bacteriostatic agents, phenoxyethanol, benzalkonium bromide, ethyl p-hydroxybenzoate and benzalkonium chloride, were detected in 20 kinds of eye drops. This method is accurate, rapid and sensitive, which could be used as a reference method for the control of bacteriostatic agents in eye drops.
