祛风湿止痛药的祛风湿药性的理论研究

Theoreitical study on the medical property of anti-rheumatic and analgesia drugs

目的 研究5味祛风湿止痛药的祛风湿药性理论。方法 采用维甲酸建立大鼠骨质疏松模型,观察祛风湿中药对骨质疏松大鼠的骨密度、抗骨折力、骨头灰化重量、骨钙素、血液生化指标Ca、P、ALP等指标;采用弗氏完全佐剂诱导大鼠建立类风湿性关节炎模型,观察其对佐剂关节炎的作用。结果 5味祛风湿止痛中药对大鼠骨密度和股骨抗折能力均有3味药达标,且能增加大鼠的骨灰分,对血清中ALP及骨钙素有3味药均达到2＋以上的作用强度;5味药均具有抑制大鼠致炎侧足肿胀的作用,降低大鼠关节炎的指数,且作用强度均达标2＋以上,显著提高了佐剂性关节炎大鼠血清中抗炎因子IL-2的水平,降低了致炎因子TNF-α的水平。结论 每味药都有自己的作用特点及侧重点,从不同的作用机制和途径起到了改善维甲酸致大鼠骨质疏松的作用及抑制大鼠佐剂关节炎的作用,改善大鼠骨质疏松及抗佐剂关节炎可能是祛风湿止痛药的药效谱之一。

OBJECTIVE To study the theory behind 5 kinds of anti - rheumatic and analgesia drugs in Chinese medicine. METHODS The model of osteoporosis of rat was established by retinoic acid to investigate the effects of anti - rheumatic and analgesia drugs on bone density, anti fracture, weight of bone ashing, osteocalcin and blood biochemistry index Ca, P, ALP. Rheumatoid arthritis rat model was induced by Freund＇ s complete adjuvant（ FCA）, and its effect on FCA arthritis was also investigated. RESULTS Influence on retinoic acid osteoporosis ： 3 of 5 kinds of anti - rheumatic and analgesia drugs were applicable to rat bone density and fracture,as well as increase bone ash. 3 drugs had 2 ＋ effects in the serum ALP and osteocalcin. Effects on the adjuvant arthritis： 5 drugs were all effective to inhibit inflammation inducing side foot swelling, decreased index of rat arthritis with the intensity of over 2 ＋ , significantly increased the level of anti inflammatory factor IL -2 in the serum of FCA arthritis rat and decreased level of inflam- matory factor TNF - α. The effect of ether extract was the strongest and the next was that of alcohol extract. CONCLUSION Every kind of drugs have its own property and emphasis. They can improve the osteoporosis of rat by retinoic acid and also inhibit rat FCA arthritis by different mode of action and route. We consider the improvement of osteoporosis by retinoic acid and inhibition of FCA arthritis of rat as one of the effect spectrums of anti - rheumatics and analgesia drugs.
