摘要
目的观察不同剂量的附子理中汤对失血性低血压过程中大鼠血压、心率、及氧化应激因子的影响。方法将实验大鼠按随机数字表法分为正常组、模型组及附子理中汤高、中、低剂量组,每组10只,除正常组外,其余4组均建立失血性低血压的大鼠模型,用多通道生理信号采集系统采集全程血压及心率;酶联免疫试剂盒检测超氧化物歧化酶(SOD)、丙二醛(MDA)、环磷酸腺苷(c AMP)的水平,观察不同浓度的附子理中汤对大鼠血压、心率等重要生命体征的改善情况。结果与正常组比较,模型组大鼠收缩压、舒张压明显下降,心率减缓;与模型组比较,附子理中汤高、中剂量组大鼠收缩压、舒张压升高(P<0.01)、心率均增快(P<0.01),低剂量组对心率无影响;模型组SOD下降,MDA与c AMP升高,附子理中汤高、中剂量组SOD上升,MDA与c AMP降低(P<0.05或P<0.01)。结论附子理中汤在一定剂量下对失血性低血压大鼠具有明显的升压、强心、调节氧化应激的作用,随着附子理中汤浓度的逐渐增加,各组药效并未见明显增强,且低剂量无效。
Objective: To investigate the effects ofFuzi Lizhong Decoction of different doses on blood pres- sure,heart rate and oxidative stress factors in rats with hemorrhagic hypotension. Methods: The rats were ran- domly divided into the nonal group,the model group,Fuzi Lizhong Decoction groups of high,medium and low dose, l0 rats in each group. Except the normal group ,the other four groups M1 establish the rat model of hemor- rhagic hypotension. The changes of whole blood pressure and heart rate were collected by multichannel physiolog- ical signal acquisition system. ELISA Kit was used to detect the level of SOD,MDA and cAMP. The improvement of the vital signs, such as blood pressure, heart rate, and so on in rats were observed with different concentration of Fuzi Lizhong Decoction. Results: Compared with the normal group,the systolic and diastolic pressure of the rats in the model group decreased and the heart rate slowed down. Compared with the model group ,the systolic and diastolic pressure increased in the high and middle dose groups (P〈 0.01),and heart rate increased fast(P〈 0.01). The low dose group had no effect on the heart rate. SOD of the model group decreased,and MDA and cAMP increased. SOD in the high and middle dose groups increased ;MDA and cAMP decreased ;the difference was statistically significant (P 〈 0.05 or P〈 0.01 ). There was no statistical significance in the low dose group. Conclusion: Fuzi Lizhong Decoction in a certain dose has a significant effect on boosting,cardiotonic and regu- lating oxidative stress in rats with hemorrhagic hypotension. It was found that with the increase of the concentra- tion of Fuzi Lizhong Decoction,the efficacy of each group was not obviously enhanced,and the low dose was inef- fective.
